Objective: In rat model of sepsis, observed at different time points rat liver myeloid differentiation protein-2 (MD2) and the expression of NF-κB of dynamic change, given exogenous TGFβ1 intervention, approaching TGFβ1 and the role of possible mechanisms in rat sepsis for the treatment of sepsis ideas.Methods:1. Grouping: 120 healthy male SD rats, weight between 220-250 g, randomly divided into three groups. The control group (sham group): 40, by 2 h, 6h, 12h, 24h and 48h further divided into five groups, each group of eight. The model group (sepsis model group): 40, by 2 h, 6h, 12h, 24h and 48h further divided into five groups, each group of eight. The establishment of cecal ligation and puncture model (CLP), the end of intravenous saline 1ml/250g after 0.5h CLP.The TGFβgroup (TGFβtreated group):40, by 2h, 6h, 12h, 24h and 48h further divided into five groups, each group of eight.The establishment of cecal ligation and puncture model (CLP), the end of intravenous injection of TGFβ20ng/ml?250g after 0.5h CLP.2.Samples taken: in various time points killing rats, sterile centrifuge tube for storage the organ,-80℃refrigerator preservation, until to reverse transcription polymerase chain reaction (RT-PCR) and Enzyme-Linked ImmunoSorbent Assay (ELISA) detection use.10% formalin solution fixed samples, at room temperature preservation, to be used pathological section HE staining.3. Results of testing: using tissue supernatant ELISA test NF-κB; RT-PCR method of detecting the expression of MD2mRNA.Results: HE staining pathology results: the control group of normal liver basic model group than the control group serious injury, infiltration of inflammatory cells, accompanied by bleeding; Compared to the control group, TGFβgroup still infiltration of inflammatory cells, but reduce infiltration of inflammatory cells and reduce the damage than the model.ELISA test results: The model group (6h,12h,24h,48h) the content increased significantly than control,(p<0.05),and 24h is the highest group,48h into the platform stage. TGFβGroup (6h, 12h, 24h) with the model group are differences (p <0.05).RT-PCR test results: MD2mRNA, a small amount of expression in the control group no difference of all time points. In the model group (6h, 12h, 24h, 48h) expression was significantly increased compared with the control group, (p<0.05), and 24h is the highest group, 48h into the platform stage. TGFβgroup (12h, 24h) expression reduction, compared with the model group are differences (p<0.05).Conclusions:MD2 in normal physiological conditions can be a small amount of expression, in septic model it is expressed significantly increased, suggesting that LPS signal transduction enhanced. Septic rat model NF-κB was increased to promote the release of inflammatory mediators, the inflammatory response is out of control. After the intravenous injection of TGFβ, MD2 expression reduced, and NF-κB levels decreased, suggesting that TGFβinhibited LPS signal transduction, reduce inflammation. But the power of TGFβin anti-inflammatory is limited. TGFβcan provide an important idea and theory for the clinical treatment of sepsis.
