Objective:To study the changes of proteomic spectra in serum from patients with laryngeal carcinoma in order to find the tumor markers which might predict the progression, metastasia and prognosis of the tumor and investigate their expression in preoperative and postoperative serum and provid a trend for curing and preventing the laryngeal cancer in the molecular biology theory.Methods:The distinct proteins in serum were detected in 32 cases of laryngeal carcinoma and 38 healthy people by IMAC-Cu proteinchip array and SELDI-TOF-MS (surface-enhanced laser desorption/ionization time-of-flight mass spectrometry).The distinct proteins in serum were detected in 32 cases of laryngeal carcinoma preoperation and within 10 days 15 postoperation with the same methods.The discriminatory profiling between preoperative and postoperative patients was analyzed by Biomarker Wizard software and Biomarker Pattern software.Results:The results show that fifteen differentially expressed proteins in serum were screened by analysis of protemic spectra of preoprative patients and normal subjects.Eight proteins(3795.04D,5068.18D,5339.78D,5908.28D,6629.74D,9266.32D,13879.6D and 14037.1D) were obtained for making up patterns that was able to class the patients-team and normal-team.Corresponding correct ratio were 87.50%(28/32)and 86.84%(33/38).Seventeen differentially expressed proteins in serum were screened by analysis of protemic spectra of preoperative and post operative patients.Six proteins(2958.52D,3796.89D,5148.86D, 5148.86D,6115.57D,4052.18D and 7770.76D) were obtained for making up patterns that was able to class the preoperative-team and postoperative-team.Corresponding correct ratio were 84.33%(56/64) and73.33%(11/15).Conclusion:It is a succeessful to develop and evaluate the different spectrometric protein profiling patterns of laryngeal carcinoma and healthy control by SELDI-TOF-MS technology.This affords possibly a new method to early diagnosis of laryngeal carcinoma.
