Objective To detect the expression of Axin andβ-catenin in human colorectal cancer and the adjacent tissue of cancer ,and their clinic pathological correlations in human colorectal cancer.Methods A total of 120 paraffin–embedded colorectal cancer and 30 adjacent tissue of cancer were studied by immunohistochemistry to evaluate the protein expression status of Axin andβ-catenin.The relation was analyzed among the protein expression and age,gender ,Dukes stage, histological classification,cell differentiation and lymphatic metastasis. And detect the relationship of the two factors'expression.Results The positive expression rate of Axin was 55.0%(66/120)in colorectal cancer ,in contrast 96.7% in adjacent tissue of cancer colorectal cancer(P<0.001).There was a significant difference inβ-catenin expression between colorectal cancer(28.3% )and adjacent tissueof cancer(93.3%)(P<0.001).In A,B,C-D,stage colorectal cancer The positive expression rate of Axin was 76.7%(23/30),55.3%(21/38),42.3%(22/52),and there was a significant difference(P=0.011). There was a significant difference between A and C-D colorectal cancer in the positive expression rate of Axin(P=0.003) .The positive expression rate of Axin was42.3%(22/52) in case with lymph node metastasis,in contrast 64.7%(44/68)in cases without lymph node metastasis(P=0.015).The nuclear or cytoplasm expression rate was 71.7%(86/120) regards as the cell membrane expression to reduce. In Well, moderately and poorly differentiated colorectalβ-catenin showd a reduced expression rate 60.0%(24/40), 65.0% (26/40) ,90.0(36/40). There was a significant difference inβ-catenin expression among well ,moderately and poorly,differentiated colorectal cancer(P=0.006). The expression ofβ-catenin in poorly differentiated colorectal was much lower than in well or moderately differentiated colorectal (P=0.002,P=0.007).Reducedβ-catenin expression rate was 82.7%(43/52) in case with lymph node metastasis,in contrast 63.2%(43/68) in cases without lymph node metastasis(P=0.019). The nuclear expression rate ofβ-catenin was 26.7%(32/120) .The positive expression rate of Axin inβ-catenin nuclear expressed colorectal cancer was 21.9%(7/32),much lower than cases without nuclear expressed 67.0% (59/88)in this study(P<0.001)。.Conclusion The expression of Axin is inversely associated with Dukes stage and lymph node positive human colorectal cancer. The reduced membranous expression ofβ-catenin is associated with poorly differentiated and lymph node positive human colorectal cancer. The expression of Axin is inversely correlated with nuclear expression rate ofβ-catenin. |
