| Object:Notochordoma is a primary malignancy of bone,which direved from the remnent tissue of brepho-chorda vertebralis.It is distributed over central axis of bodies,with growing slowly,infiltrative growth.There is a slow metastasizing with notochordoma.MMP-13 is one member of the metalloproteinase family.It can almost degrade all of extracellarma trix and basal membrane.Its main substrate includes collagen fibersâ… ,â…¡,â…¢,â…£,gelatinum,laminin and aggrecan.At present,we found the espression of MMP-13 in many sorts of tumors.It plays an important role in the process of tumors infestation and metastasis.MMP-14 is one of membrane-type matrix metalloproteinase,which belongs to metalloproteinase family.It also can be called MT1-MMP.MMP-14 is located on cell envelope.Its main substrate includes collagen fibersâ… ,â…¡,â…¢,anchylo-fiber protein,dermatan sulfatase proteoglycan,laminin-â… and vitronectin.at present,there are many reports about more expression in tumor tissue such as gastric cancer,breast cancer,bronchogenic carcino -ma,colon carcinoma,encephaloma,liver cancer than in innocuousness tissue.The expression of MMP-14 can promote invasive growth of tumor.Osteopontin is a phosphorylated protein.it can be found at many sorts of tissue,such as kidney,auris interna,caduca,cake uterine,smooth muscle,T cell,macrophagus.In recent years,the study shows there are high expressions of Osteopontin in gastric cancer,breast cancer,bronchogenic carcinoma,liver cancer,esophageal carcinoma,prostatic carcinoma. High expressions of Osteopontin have close relation with capacity of tumor cells's metastasis and the growth of tumor blood vessel.Cyclooxygenase is a rate-limiting enzyme in the process of prostaglandin synthesis. There are Cyclooxygenase-1 and Cyclooxygenase-2 in mammalian.in recent decade,many studies show Cyclooxygenase-2 play an important role in tumor origin and development.the functin of Cyclooxygenase-2 include influencing generation and differentiation of cells,restraining apoptosis;promoting the growth of tumor blood vessel,enhancing capacity of tumor cells's metastasis;restraining immune reaction.In our studies,we detect the expression of MMP-13,MMP-14,Osteopontin, Cyclooxygenase-2 so that to study the meaning of origining and developing in notochordoma.it can explain etiopathogenisis of notochordoma from multiple aspects and give new thinking for diagnosis and treatment of notochordoma.Methods:Collecting notochordoma specimens and innocuousness tissue specimens. Constructing one tissue arrayer,which contain 94 points.Immunohistochemistry was applied to detect the expression of MMP-13,MMP-14,Osteopontin,Cyclooxygenase-2.to observe the relationship among them.To further observe the relation between the expression of them and the associated clinicopathological features.Log-rank test were used for signal-factor analysis with the expression of MMP-13,MMP-14 and Osteopontin. Kaplan-Meier method was used for making the survival curve.Results:1,Constructing one tissue arrayer of 94 points,which contains 75 points of notochordoma and 19 points of peritumor tissue.2,The positive expression of MMP-13,Osteopontin protein colors cytomembrane and cytolymph with pale yellow,dark yellow or brown.The positive expression of MMP-14 protein colors cytomembrane with pale yellow,dark yellow or brown.The positive expression of MMP-14,MMP-13,Osteopontin by IHC were 78.67%,81.33%,61.33%, respectively.The positive expression of COX-2 protein colors cytolymph and caryotheca with pale yellow,dark yellow.The positive expression of COX-2 by IHC was 4%.The expressions of MMP-14,MMP-13,Osteopontin between notochordoma and non-tumor have a remarked difference(P<0.05).The expressions of COX-2 between notochordoma and non-tumor have no difference(P>0.05).3,The expression of MMP-13 and MMP-14 had a closely relation in notochordoma (P<0.01).The expression of MMP-13 and Osteopontin had a closely relation in notochordoma(0.01<p<0.05).The expression of Osteopontin and MMP-14 had a closely relation in notochordoma(0.01<p<0.05).The expression of COX-2 and MMP-13,MMP-14,Osteopontin had no relation in notochordoma(P>0.05).4,The expression of MMP-13 had no relation with patient's sex,age,tuomr location and vertebral quantity in notochordoma(P>0.05).The expression of MMP-14 had no relation with patient's sex,tuomr location and vertebral quantity in notochordoma(P>0.05).The expression of MMP-14 had a closely relation with patient's age in notochordoma (P<0.01).The expression of Osteopontin had no relation with patient's sex,age,tuomr location and vertebral quantity in notochordoma(P>0.05).5,There are significance difference between non-tumor survival periods of the positive expression group of MMP-13,MMP-14 and the negative expression group through Log-rank tests(P<0.05),The positive expression group of Osteopontin had no significance difference with the negative sufferers(P>0.05).Conclusions:1,The level of three proteins in notochordoma tissue are more high than its in non-tumor tissue.2,The expression of MMP-13,MMP-14 and Osteopontin has a closely relation in notochordoma.the correlation among them lead to malignant activity of notochordoma.3,The level of MMP-13,MMP-14 and Osteopontin had no relation with patient's sex, tuomr location and vertebral quantity in notochordoma.The expression of MMP-14 had a closely relation with patient's age in notochordoma.the amount of MMP-14 is rising as the patients are aging.The amount of MMP-14,MMP-13 and Osteopontin also do it.4,MMP-13,MMP-14 proteins are important factors influencing CRC prognosis, Non-tumor survival periods of the positive expression of MMP-13,MMP-14 is less than that of tha negative expression of MMP-13,MMP-14,Significance correlation occurred between the different levels of MMP-13,MMP-14 and recurrence of the spinal chordoma. |
