Objective To determine the population structure of genotypes and Mate-typing of 115 clinical Cryptococcus neoformans species complex isolates from Chinese HIV-Positive and HIV-Negative patients in Southeast China by ITS gene,CAP59 gene sequence analyses and the M13-PCR fingerprinting survey.Methods A total of 115 Chinese clinical Cryptococcus neoformans species complex isolates were analysed by sequences of Capsule59 and ITS gene using CLUSTAL W1.83 and MEGA3.1 soft,phylogenetic tree which statistical tested by bootstrapping method was proformed,and genotyped analysis was used PCR-fingerprinting by the M13 oligonucleotide as a single primer,mating-type was determined by PCR using two specific primer pairs in accordance with Chaturvedi et al.Results 1.VNⅠ(76.5%) was the most represented genotypes,followed by VNⅡ(14.8%),VNⅢ(2.6%),VGⅠ(4.3%), VGⅡ(0.87%) and VNⅣ(0.87%) of 115 clinical Cryptococcus neoformans species complex isolates from Chinese HIV-Negative and -Positive patients;2.The mating type determinations revealedαis the most represented mating-type(97.4%) of 115 clinical Cryptococcus neoformans species complex isolates from Chinese HIV-Positive and -Negative patients.3.The 6 Cryptococcus neoformans var.gattii strains were mainly isolated from patients of Shanghai,South regions of Jiangsu and Zhejiang Province.4.Serotypes can not identified only by analysis of CAP59 gene sequence,however,ITS sequencing is able to identify clinical isolates on variety level. Conclusion 1.The isolates of mate-typeα,VNI genotype is the most majority of clinical Cryptococcus neoformans species complex isolates from Chinese HIV-Positive and HIV-Negative patients,however,1 isolates of mate-typeα/A,VNⅢgenotype,were isolated from 20 Chinese AIDS patients.2.Genetic polymorphisms of the Chinese clinical isolates of Cryptococcus neoformans var.gattii was also observed in this study,and the Chinese clinical Cryptococcus gattii strains are mainly isolated from Chinese subtropical regions.3.The mating-typeαaccount for 97.4%of the clinical Cryptococcus neoformans species complex isolates from Chinese HIV-Positive and HIV-Negative patients.4.Sequences of ITS and CAP59 genes analyses is able to classified Cryptococcus neoformans strains on variety level and our results furnish new information on the molecular epidemiology of Cryptococcus neoformans isolates from Southeast China.
