Objective: To investigate the effect of angiotensinâ…¡and angiotensinâ…¡receptor antagonist candesartan on matrine on proliferation of rat Hepatic Stellate Cells line-99(rHSCs-99) in vitro and the expression of monocyte chemoattractant protein-1(MCP-1).Methods: HSC line rHSCs-99 were incubated in vitro. After subcultivation, rHSCs-99 were divided into four groups:â‘ Control group: only administration of 1% FBS medium ;â‘¡Angiotensinâ…¡group: 1% FBS medium plus angiotensinâ…¡of final concentration 10-10, 1O-9, 1O-8, 1O-7 and 1O-6 mol/L separately;â‘¢Candesartan group: 1% FBS medium plus candesartan of final concentration 10-10, 1O-9, 1O-8, 1O-7 and 1O-6 mol/L separately;â‘£Candesartan plus angiotensinâ…¡group: administration with candesartan of final concentration 1O-9, 1O-8, 1O-7 and 1O-6 mol/L separately in advance, angiotensinâ…¡of final concentration 1O-7 mol/L was added 30 minutes later. After intervention of drugs 24 hours, MTT assay was taken to test the proliferation of rHSCs-99. After intervention of drugs 36 hours, enzyme-linked immunosorbent assay (ELISA) was taken to test the content of MCP-1 in medium of rHSCs-99. The results were analyzed with SPSS13.0 statistics soft.Results:1. Angiotensinâ…¡group: The result of MTT assay showed that 1O-9, 1O-8, 1O-7 and 1O-6 mol/L angiotensinâ…¡separately can promote the proliferation of rHSCs-99, compared with control(P<0.05). The result of ELISA showed that 10-10, 1O-9, 1O-8, 1O-7 and 1O-6 mol/L angiotensinâ…¡separately can increase the content of MCP-1 in medium of rHSCs-99, compared with control(P<0.05).In the scope of experimental concentration angiotensinâ…¡can promote the MCP-1 secretion of rHSCs-99 by dose-dependent manner(r=0.936,p=0.000). 2. Candesartan group: The result of MTT assay showed that 1O-8, 1O-7 and 1O-6 mol/L candesartan separately can inhibit the proliferation of rHSCs-99, compared with control(P<0.05). The result of ELISA showed that the content of MCP-1 in medium of rHSCs-99 in 10-10, 1O-9, 1O-8, 1O-7 and 1O-6 mol/L candesartan separately was no significant change, compared with control(P>0.05).3. Candesartan plus angiotensinâ…¡group: The result of MTT assay showed that the experiment group of administration with 1O-9, 1O-8 and 1O-7 mol/L candesartan separately in advance can also promote the proliferation of rHSCs-99, compared with control(P<0.05).The experiment group of administration with 1O-6 and 1O-5 mol/L candesartan separately in advance can not promote the proliferation of rHSCs-99,compared with control(P>0.05). The result of ELISA showed that the experiment group of administration with 1O-9 and 1O-8 mol/L candesartan separately in advance can also increase the content of MCP-1 in medium of rHSCs-99, compared with control(P<0.05). The content of MCP-1 in medium of rHSCs-99 in these groups was no significant change, compared with 1O-7 mol/L angiotensinâ…¡group(P>0.05). The content of MCP-1 in medium of rHSCs-99 in the experiment group of administration with 1O-7 , 10-6 and 10-5 mol/L candesartan separately in advance decreased significantly, compared with 1O-7 mol/L angiotensinâ…¡group(P>0.05).Conclusions: Angiotensinâ…¡can significantly stimulate the proliferation of rHSCs-99 and the synthesis and secretion of MCP-1. Angiotensinâ…¡receptor antagonist candesartan can inhibit the proliferation of rHSCs-99. It can inhibit the effect of increasing the synthesis and secretion of MCP-1 with administration of angiotensinâ…¡.
