| Objective:To investigate the roles of the PI3K-Akt signal in the proliferation and MMP-2,MMP-9 expression of gastric carcinoma cell HGC-27, the specific PI3K inhibitor LY294002 was taken to the cells,inhibiting the activation of the PI3K-Akt signal.Methods : The HGC-27 cells were incubated with different concentrations(0,10,20 and 40μM separately) of LY294002 for different times(20,40 and 60hours separately);CCK-8 was taken to test the cell proliferation; Propidium Iodide dying was taken to test the dislocation of cell cycle;immunocytochemistory was taken to test the expression of p-Akt ;semi-quantity RT-PCR was taken to test the expressions of mRNA of MMP-2 and MMP-9.Results: When the HGC-27 cells were treated with LY294002:1.the cell proliferation was inhibited(P<0.05),and within the experiment span,the effect of proliferation inhibition was increased with the concentrions(P<0.05) of LY294002 and interfering times(P<0.05);2.p-Akt expression was suppressed;3.the cell cycle analysis showed that the rate of S stage cells was decreased(P=0.008);4.the expression of mRNA of MMP-9 was decreased(P=0.000),and meanwhile the expression of mRNA of MMP-2 was not obviously interfered(P=0.118).Conclusions:1.The activation of PI3K-Akt signal is inhibited after the HGC-27 cells are treated with LY294002;2.LY294002 can inhibit the proliferation and the cell cycle progression of HGC-27 cells;3.PI3K-Akt signal takes part in modulating the MMP-9 expression of HGC-27 cells,and the expression can be inhibitted by suppressing the activation of the signal. |
