Objective: Chronic atrial fibrillation (AF) is characterized by a remodeling process which involves the development of fibrosis. Basic fibroblast growth factor( bFGF) has been suspected to be involved in this process . To provide laboratory evidences, this work has designed to observe the impact of bFGF on the collagen synthesis in cultured cardiac fibroblast(CFs) of human. Whether bFGF had acted as neuroendocrine mediator through non-hemodynamic manner on myocardial ,a pathologic process that was seen in AF were then discussed.Methods:CFs of human were isolated by trypsin digestion method and the cell culture systems were established.And the expression of collagen type I mRNA was detected by reverse transcription polymerase chain reaction( RT-PCR ).Results:(l)Highly enriched fibroblast cultures were obtained as measured by Immunohistochemical staining vimentin Positive rate was>98%; (2) collagen type I mRNA levels of the cells Treated with bFGF were significantly higher than that of the control (all,P<0.05).Conelusions:BFGF significantly inerease the collagen type I mRNA cexpression, Characterized in a concentration-dependent manner .From this work we proposed that bFGF could be another potent activator causing cardiac fibrosis,independent of other hemodynamic Regulators and,with other neuroendocrine mediators,bFGF may jointly takes Part in the development of myocardial remodeling.
