Objective to investigate the relationship between the variance of Th cell subsets and the expression of TLR2 mRNA in peripheral blood monocytes of patients with chronic HBV infection .Methods Expression of TLR2 mRNA in PBMC was determined by RT-PCR, the levels of Th cell subsets were measured by flow cytometry.Results①The percentages of Th1 cell in HBeAg(+) group and HBV-DNA replicating group were significantly lower than that of control group(9.42±2.03,9.52±2.41 vs 11.18±3.12,P <0.05). The percentages of Th2 cell in HBeAg(+) group ,ALT increasing group,HBV-DNA replicating group were significantly higher than that of control group(3.05±1.12,3.12±1.12,2.97±1.12 vs 2.38±0.96,P <0.05). All the Th1/Th2 ratios of HBeAg(+) group, ALT increasing group and HBV-DNA replicating group were significantly lower than that of control group(P <0.05).②It was confirmed that TLR2 mRNA could be detected in PBMC of human. No significant differences were detected in TLR2 mRNA between CHB and control.The levels of Th1,Th2 were not correlated with the expression of TLR2 mRNA in PBMC.Conclusion①The results suggest that Th1/Th2 ratio decrease and Th2 cell increase in the patients with chronic HBV infcetion, the disturbance of Th1/Th2 is associated with hepatic injury and HBV replication . The Th1/Th2 ratio can be taken as an important index to moniter the immune state of patients with HBV infection.②The results indicate that TLR2 may not play a role in liver injury and the expression of TLR2 mRNA in PBMC mignt not be correlated with variance of Th cell subsets.
