NGF Associated With DDP On Apoptosis In Human Lung Adenocarcinoma Cell Lines A549

Objective To evaluate the effects of NGF associated with DDP on apoptosis in human lung adenocarcinoma A549 cell lines.Methods The A549 cell lines were cultured in saturated humidity incubator with RPMI-1640,37℃,5%CO₂, which were randomly divided into normal control group(A549 cells,PRMI-1640),DDP group(A549 cells,PRMI-1640,DDP),NGF group (A549 cells, PRMI-1640,NGF5,10,15 ug/ml), NGF+DDP group(A549 cells, RPMI -1640,NGF10ug/ml and DDP5ug/ml).The cultured cells were collected after 48 hours, then MTT was used to evalue the inhibition ratio of A549 cells,electron microscope (TEM) was used to observe the shape of the cells and flow cytometry was used to detect the rate of apoptosis as well.Results The inhibition ratio of A549 cells observed with MTT is that NGF can inhibite the growths of A549 cells. Increasing the concentration of NGF,the inhibition ratio would be going up. The inhibition ratio of NGF,DDP,NGF associated with DDP groups were higher than the normal group. The inhibition ratio of NGF associated with DDP group was higher than others. Comparison had statistical significance (P<0.05). The karyopyknosis,chromatin enrich, apoptotic body was observed under TEM in DDP group,NGF group,NGF associated with DDP group after 48h while cell nucleusthe,mitochondrion and endoplasmic reticulum were clear in normal control group,nucleus held the most of the cell. Cellular shape held normal in normal control group. DDP group,NGF group and NGF associated with DDP group had markedly increased in apoptosis rate than normal control group (P<0.05),NGF associated with DDP group had significant difference in apoptosis rate with DDP group,NGF group (P<0.05), while NGF(5ug/ml,10ug/ml) groups and DDP group had no significant difference (P > 0.05).Conclusions NGF associated with DDP can remarkbably induce the cellular apoptosis.
