| Aim: To establish a stable and strong repeatable heart failure rat model, to estimate its heart function's changing by ultrasonic echocardiogram and to evaluate the feasibility of ultrasonic echocardiogram's application on rat model.Methods: A total of 120 2-month-old male SD rats weighing 200-220g were randomly assigned to 2 groups: operation group, including 100 rats, recieved a tnoracoter: open the arcula cordis, deligate the anterior descending branch of left arcula cordis; sham operation group, including 10 rats, received the tnoracoter and open the arcula cordis as the same, but did not deligate the anterior descending branch of left arcula cordis; normal group, including 10 rats, without any management .To observe the change of the rats in mental status, action status, fur, diet, and breathing rate every day after the operation. For appraising the change of weight form start to the end, we measured the rats' weight one day before surgery and the end of the 6th week after the surgery. For appraising the change of heart function of heart failure rats, we gave the echocardiography on the end of the 6th week to all the rats. We get the blood serum to detection the level of brain natriuretic peptide (BNP) on the end of the 6th week. Result: Operation group which was survived appear obviously objective sign of heart failure about 10 days later after the surgery, as listlessness, increasing of blepharon excretion, descending of locomotor activity, depilating, accelerated breathing, temperature drop, and to be fond of get together, et, al.The weight of the normal group and the sham operation group was increased obviously, but the weight of the heart failure group was increased slowly. There is difference between the last weights of the three groups (P<0.05) .The heart function of the operation group was obviously lower than the normal group and the sham operation group, it was showed as left ventricle exponent (LV exponent) get higher, left ventricle posterior wall exponent (LVPW exponent) and interventricular septum exponent (IVS exponent) get lower, fraction shortening (FS) and ejection fraction (EF) get lower (P<0.05) . There is no difference in left atrium exponent (LA exponent) (P>0.05).The blood-serum brain natriuretic peptide (BNP) level in operation group was obviously higher than that in normal group and sham operation group, and the degree of its advance offered a linear relationship with the degree of heart function's decline( including raise of the LV exponent, decline of the IVS exponent, LVPW exponent, EF and FS ).We confirmed the modeling was succeed on aspects of objective sign and ethology, the difference between first weight and last weight, the changing of heart function and the changing of BNP level. Ultrasonic echocardiogram can evaluate the change of heart function on heart failurerats well, and it is feasible and reliable.Conclusion: To deligate the anterior descending branch of left arcula cordis of rats can make heart failure model successfully. At the same time, ultrasonic echocardiogram can evaluate the heart function's changing on heart failure rat model well, and the indicatrix of it offered a linear relationship with the BNP level of rat. |
