| Objective:1. To establish Mongolian Gerbil model with Hp infectious gastritis by inoculated with Hp SS1 after antibiotic pretreatment.2. To research the efficacy of Bacillus acidi lactici CMS002 strain and combine with PPI treatment to Hp infectious gastritis and its influence to bacterial numbers in stomach.3. Observe and compare the expression of NF-κB and IL-1β, TNF-αto explore the role of activation of NF-κB in the pathological mechanism of Hp infectious gastritis and the mechanism of Bacillus acidi lactici CMS002 strain treatment to Hp infectious gastritis.Methods: To establish Mongolian Gerbil model with Hp infectious gastritis by inoculated with Hp SS1 after antibiotic pretreatment, a total of 32 Mongolian Gerbil model were randomly divided into 4 groups, 8 in each group: Group1 was treated with PPI and antibiotics,Group2 was treated with Bacillus acidi lactici CMS002 strain,Group3 was treated with PPI and Bacillus acidi lactici CMS002 strain and Group4 was treated with sodium chloride solution. All the four groups were treated for 7-10 days respectively. Additionally, 8 Mongolian Gerbil in group5 were normal, without any treatment. All experimental mice were sacrificed 4 weeks post-treatment. To observe and compare the general condition,the infection and eradiation of Hp,the pathohistologic changes,the changes of bacterial numbers in stomach,the expression of NF-κB and IL-1β, TNF-αof each group.Results:1. Results of the establishment of Mongolian Gerbil model: In Mongolian Gerbil model, Hp colonization rate were 100% and there were a lot of lymphoid cells congregated.2. The infection and eradiation of Hp: In group1, Hp infection rate and Hp eradiation rate were 12.5% and 87.5% respectively; 25% and 75% in group2; 25% and 75% in group3; 100% and 0 in group4; Hp infection rate was 0 ingroup5. Compared with group4, Hp infection rate and Hp eradiation rate in group1,group2 and group3 was significantly decreased and increased seperately (P<0.05), there was no significant difference in group2,group3 and group1 (P>0.05 ).3. The pathohistologic changes: Compared with group5 and group4, the scores in the chronic inflammation of gastric mucosa of group1,group2 and group3 were significantly increased and decreased seperately(P<0.05), there was no significant difference in group2,group3 and group1 (P>0.05).4. The changes of bacterial numbers in stomach: Compared with group5, the bacterial numbers in stomach of group4 and group1 were significantly decreased (P<0.05). Compared with group5, the bacterial numbers and the numbers of Bacillus acidi lactici in stomach of group2 and group3 were significantly increased (P<0.05), the numbers of Bacillus bifidus and Bacteroid in stomach of group2 were significantly decreased(P<0.05), there was no significant difference in group2,group4 and group1 (P>0.05), compared with group4 and group1, the numbers of Bacillus bifidus and Bacteroid in stomach of group3 were significantly increased (P<0.05), there was no significant difference between group3 and group5o No Staphylococcus aureus was found in stomach of each group.5. The expression of NF-κB and IL-1β, TNF-α: Compared with group5 and group4, the scores of NF-κB and IL-1β, TNF-αof group1,group2 and group3 were significantly increased and decreased seperately (P<0.05), there was no significant difference in group2,group3 and group1 (P>0.05).Conclusions:1. Mongolian Gerbil model with Hp infectious gastritis was successfully established by inoculated with Hp SS1 after antibiotic pretreatment in our study.2. Bacillus acidi lactici CMS002 strain could inhibit the growth of Hp efficiently, its efficacy could enhance when combined with PPL3. The pathological mechanism of Hp infectious gastritis probably focused on the activation of NF-κB, one of the mechanism of Bacillus acidi lactici CMS002 strain treatment to Hp infectious gastritis probably was concerned with the block of the NF-κB message route. |
