Objective:randomization test to examine the expression of DNMT1 ans DNMT3a and DNMT3b in eutopic and ectopic endometrium of women with endometriosis and compared the expression levels with that in the endometrium of women without endometriosis ,to study the role of DNA methyltransferase in the mechanism of the endometriosisMethod: RT-PCR ,Real Time RT-PCR were used to examine the expression of DNMT1,DNMT3a ,DNMT3b in the in eutopic and ectopic endometrium of 20 women with endometriosis and compared the expression levels with that in the endometrium of 20 women without endometriosis , and immunofluorescene staining was used to detect the expression of DNMT1 in these tissues.Result: the expression levels of DNMTI, DNMT3a, DNMT3b were significantly lower in the ectopic endometrium and eutopic endometrium as compared with control subjects, P<0.05,the results have statistically significant ,The fold changes in ectopic endometium as compared with control subjects were 0.44 , 0.12 and 0.27 for DNMT1,NMT3a, and DNMT3b,respectively. For eutopic endometrium ,the corresponding fold changes were 0.27, 0.13, 0.15. But the expression levels of DNMT1, DNMT3a, and DNMT3b were similar in ectopic endometrium and eutopic endometrium, P>0. 05. Conclusion:the aberrant expression of DNMT1 and DNMT3a and DNMT3b in the women with endometriosis may be play a role in the mechanism of endometriosis ,This provides another piece of evidence that endometriosis may be ultimately an epigenetic disease...
