| Object:To approach the therapeutical effect of L.acdophilus CMS-H002 on acute ulcerative colitis(UC)model,and study the expression variance of TLR4,NF-κBp65,MCP-1 in mice colonic tissue,as well as provide experiment foundation for the therapy of Lactobacillus acidophilus on UC.Methods:1.Establish mice acute UC model:Colitis was induced by exposing BALB/c mice to 2.5%DSS(MW: 50000)in drinking water for 7 days.2.Investigate the effect of L.acdophilus CMS-H002 strain on acute UC model:The BALB/c mice were randomly divided into five groups:nomal control group,NS control group,salicylazosulfapyridine(SASP)group, high dose L.acdophilus CMS-H002 strain group,and low dose L.acdophilus CMS-H002 strain group.Colitis was induced by exposing BALB/c mice to 2.5%DSS in drinking water for 7 days.The nomal control group did not accept any treatment.NS group was infused with 0.4ml 0.9%NaCl per day.The positive group was infused with SASP(15mg/10g)per day.Two CMS-H002 groups in doses of 4.0×10~8 cfu/10g,4.0×10~7 cfu/10g were administered orally once a day individually,beginning concurrently with exposure to DSS.The body weight and the character of feces as well as occult blood were observed to know disease activity index of every mouse per day.Seven days later,all mice were sacrificed.The colon length was measured and the colony tissue was handled with HE stained.3.Detect the expression of Proteins TLR4 and NF-κBp65 in the colonic mucosa by immunohistochemical staining,and MCP-1mRNA by semi-quantatitive reserve transcriptional(RT)PCR.Results1.NS group mice were hair-withered,depressed,sluggish, weight-loss,diarrheic,and even bloody stools,the body weight lose obviously,to the day 7,it had reduced 3.05±0.70.After L.acdophilus CMS-H002 strain and SASP treatment,body weight loss,diarrhea and bloody stools were decented.The high dose of L.acdophilus CMS-H002 strain was found more effective on protecting weight loss.2.The DAI scores of NS group mice increased from 0 to 6.53±1.50, and decreased after treated by L.acdophilus CMS-H002 strain and SASP. To the day 7,the scores of the high dose group was 1.5±0.74,lower than 2.3±0.83 of the SASP group,but there was no statistical significance between them(P>0.05).3.The length of colon reduced obviously in NS group than that in nomal group,and the mucosa became congestive and dropsical,the tissue under microscope showed incompleteness of mucosa,loss of goblet cells and destruction of crypt,hemorrhage of mucosa and infiltration of inflammation cells.After treated with L.acdophilus CMS-H002 strain and SASP,the decurtation of colon,the loss of epithelium and the inflammation was prevented.And the histology scores of the high dose group was 4.7±0.90,lower than 6.6±1.60 of NS group(P<0.05).4.The level of TLR4,NF-κBp65,MCP-1 mRNA in colon tissue of NS group increased obviously.It reduced after treated with L.acdophilus CMS-H002 strain and SASP,and the high dose group was the lowest.5.It existed positive correlation between The level of TLR4,NF-κBp65,MCP-1 mRNA and DAI scores/histological scores,r>0.398, P<0.01. Conclusions1.The UC model induced by 2.5%DSS showed features of acute stage of UC with low mortality and could be used in research of mild and moderate UC.2.The L.acdophilus CMS-H002 strain can be used to treat acute UC induced by DSS effectively,the effect to prevent inflammation may be related to some component or some metabolite of the stain,that can prevent the MCP-1 expression by TLR4/NF-κB pathway. |
