Objective:To explore the mechanism of estrogen on delaying skin aging by observing the expression of matrix metalloproteinase-1 gene in human dermal fibroblasts in vitro.Methods:Human skin fibroblast were cultured in vitro.These fibroblast cells were then divided into 3 groups,in which no estrogen was added in the medium in the control group,lower dimensional estrogen was added in the medium in the second group,higher dimensional estrogen added in the medium in the third group.A few days later,we collected the fibroblast,which was index number growth phase from different groups,extracted RNA,then amplified by RT-PCR.The PCR product was determined by agar gum electrophoresis,to analysis the level of MMP-1 mRNA.Results:Human skin fibroblast proliferates well in vitro,taking on Fusiform or babylon weeping shape.Growth curve showed latency phase,index number growth phase and platform phase,we also found latency phase was shorter,about 0 to 2 days,then index number growth phase.After about 5 days fibroblast cells proliferates stable.Agar gum electrophoresis showed,there was no significant differences in the level of MMP-1 mRNA between the low-dose group and the control group.However, the level of MMP-1 mRNA was lower in the high- dose group compared to control group.There was significant differences(P<0.05).Conclusion Estrogen at high concentration exhibites inhibitory effects on mRNA expression of matrix metalloproteinase-1,but low concentration not.
