Objective:Survivin gene plays an important role in suppressoring cell apoptosis.CyclinD1 takes the role as enhancing the cell proliferation.We aim to determine the expression of Survivin and CyclinD1 in epithelial ovarian tumors and to study the relationship of their expression with clinicpathological characteristics and to demonstrate the potential use of them as a diagnostic and therapeutic agent.Methods:The expression of Survivin and CyclinD1 in 20 of normal ovarian tissue, 15 of benign ovarian tumors,40 of epithelial ovarian carcinoma were examined by S-P immunohistochemistry. The level of CyclinD1 and Survivin in 40 ovarian carcinoma was analyzed against clinicopathological factors such as clinical stage and histological grade, morover, CyclinD1 expression in comparison with Survivin expression was investigated preliminarily.Results:1.The percentage of Survivin positively expression decreased from ovarian carcinoma(67.5%)to benign ovarian tumors(6.7%)to normal ovarian tissue(0%).Among them,there were statistically significant between ovarian carcinoma and benign ovarian tumors(p<0.05)and between ovarian carcinoma and normal ovarian tissue(p<0.05).But no statistically difference was found between benign ovarian tumors and normal ovarian tissue(p>0.05).2.In ovarian cancer group,the expression of Survivin in FIGO III~IV stage or lymph node metastasis group or bad differentiated group was significantly higher than in FIGO I~II stage or no lymph node metastasis group or well differentiated group(p<0.05).The expression of Survivin in serous carcinoma group or age older than 50 years old group was higher than in non-serous carcinoma or age younger than 50 years old group,but the difference was not significant(p>0.05).3.The expression of CyclinD1 was respectively decreased in ovarian carcinoma ,benign ovarian tumors,normal ovarian tissue. The positive expression was statistically difference between ovarian carcinoma and benign ovarian tumors(p<0.05)and between ovarian carcinoma and normal ovarian tissue(p<0.05).However,there were no statistically difference between benign ovarian tumors and normal ovarian tissue(p>0.05).4.In ovarian cancer group,the expression of CyclinD1 in lymph node metastasis group or bad differentiated group was significantly higher than no lymph node metastasis group or well differentiated group(p<0.05).No correlation was found between the expression and age ,FIGO stage,histological subtype.5.In ovarian cancer group,there was positive correlation between the expression of Survivin and expression of CyclinD1.Conclusions:1 .Elevated expression of Survivin was observed in ovarian carcinoma,compared with benign ovarian tumors.There was no expression of Survivin in normal ovarian tissue.It suggested that expression of Survivin may play a role in the carcinogenesis of ovarian carcinoma.2.The expression of Survivin was correlated with FIGO stage,histological grade, lymph node metastasis and had no correlation with age, histological subtype.It suggested that expression of Survivin may make it easier for epithelial ovarian carcinoma to metastases, which cause poor prognosis.3.Elevated expression of CyclinD1 was observed in ovarian carcinoma,compared with benign ovarian tumors and normal ovarian tissue.It suggested that expression of CyclinD1 may play a role in the carcinogenesis of ovarian carcinoma.4.The expression of CyclinD1 was correlated with histological grade,lymph node metastasis and had no correlation with FIGO stage,age,histological subtype.It suggested that expression of CyclinD1 may make it easier for epithelial ovarian carcinoma to metastases,which cause poor prognosis.5.In epithelial ovarian carcinoma group,expression of Survivin had a strong correlation with expression of CyclinD1.It suggested that Survivin may be in coordinated with CyclinD1 in carcinogenesis and progression of epithelial ovarian carcinoma.
