Objective: To detect the expression of Survivin,VEGF-C,CyclinD1,CDC25A and PCNA in cholangioncarcinoma tissue, explore their mechanism, search for new therapy of cholangioncarcinoma.Methods1 Researching 42 cases of cholangioncarcinoma tissue,17 cases of adjacent tissue to carcinoma and 20 cases of normal tissue1.1 Immunohistochemical technique (SP) was used to examine the expression of Survivin,VEGF-C and PCNA in 42 cases of cholangioncarcinoma tissue and 20 cases of normal tissue. The expression degrees were analyzed relating to clinicopathological features of the patient including age, sex, loci of the tumor, histology type, lymph node metastasis etc. All specimens were diagnosed by pathologist.1.2 To examine the expression degrees of CyclinD1,CDC25A in 42 cases of example cholangioncarcinoma tissue and 20 cases of normal tissue by FCM, research the expression degrees of the protein. All specimens were diagnosed by pathologist. 1.3 To examine the mRNA of Survivin,VEGF-C,CyclinD1,CDC25A and PCNA in 42 cases of cholangioncarci- noma tissue,17cases of paired adjacent tissue to carcinoma and 20 cases of normal tissue by RT-PCR. The examples were all diagnosed by pathology.2 Making use of the SAS Systerm for Windows v6.12 software carry out statistical treatment;Using x±s express Test data; using ANOVA compare in different groups;Usingχ2 detect the result of immunohistochemical technique (SP);statistical significance is considered whenα=0.05.Results1 The expression of protein of Survivin,VEGF-C,PCNA in all cases of different pathological change of bile duct tissue detected by immunohistochemical technique SP.The protein expression of Survivin in cholangioncarcinoma tissue and normal bile duct tissue was 78.57% and 25.00% respectively, the protein expression of Survivin was higher in bile duct carcinoma tissue than normal bile duct tissue (P<0.05); The protern expression of VEGF-C in cholangioncarcinoma tissue and normal bile duct tissue was 83.33% and 30.00% respectively, the protein expression of VEGF-C was higher in bile duct carcinoma tissue than normal bile duct tissue (P<0.05); The protein expression of PCNA in cholangioncarcinoma tissue and normal bile duct tissue was 71.19% and 25.00% respectively, the protein expression of PCNA was also higher in bile duct carcinoma tissue than normal bile duct tissue. (P<0.05) 2 The expression of protein of CyclinD1 and CDC25A in all cases of different pathological change of bile duct tissue detected by FCMThe protein expression of CyclinD1 cholangioncarc- inoma tissue and normal bile duct tissue was 530.253±27.823 and 497.416±38.019 respectively, the protein expression of CyclinD1 was higher in bile duct carcinoma tissue than normal bile duct tissue (P<0.05); The protein expression of CDC25A in cholangioncarcinoma tissue and normal bile duct tissue was 534.029±35.674 and 497.276±42.343 respectively, the protein expression of CDC25A was also higher in bile duct carcinoma tissue than in normal bile duct tissue (P<0.05).3 The expression of Survivin,VEGF-C,CyclinD1,CDC25A and PCNA mRNA in all cases of different pathological changes of bile duct tissue.The expression of Survivin mRNA in cholangioncarcinoma tissue,adjacent tissue to carcinoma and normal bile duct tissue respectively was 0.6238±0.080,0.5693±0.0443,0.5684±0.0317; The expression of Survivin mRNA was higher in bile duct carcinoma than in the others(P<0.05).The expression of VEGF-C mRNA in cholangioncarcinoma tissue,adjacent tissue to carcinoma and normal bile duct tissue was 0.6930±0.1078,0.6270±0.0664,0.6105±0.0577 respectively; The expression of VEGF-C mRNA was higher in bile duct carcinoma tissue than in the others(P<0.05). The expression of CyclinD1 mRNA in cholangioncarcinoma tissue,adjacent tissue to carcinoma and normal bile duct tissue was 0.6288±0.0942,0.5608±0.0577,0.5845±0.0310 respectively; the expression of CyclinD1 mRNA was higher in bile duct carcinoma tissue than in the others(P<0.05).The expression of CDC25A mRNA in cholangioncarcinoma tissue,adjacent tissue to carcinoma and normal bile duct tissue was 0.5937±0.0716,0.5229±0.0575,0.5135±0.0780 respectively; the expression of CDC25A mRNA was higher in bile duct carcinoma tissue than in the others(P<0.05); The expression of PCNA mRNA in cholangioncarcinoma tissue,adjacent tissue to carcinoma and normal bile duct tissue was 0.6303±0.0773,0.5692±0.0408,0.5605±0.0331 respectively; the expression of PCNA mRNA was also higher in bile duct carcinoma tissue than in the others(P<0.05).ConclusionThe expression degree of protein was accordant with that of mRNA on Survivin,VEGF-C,CyclinD1,CDC25A and PCNA,all of them were higher in cholangioncarcinoma tissue than in normal bile duct tissue.Through the changes of these gene and protein expression, maybe it is possible to elucidate the carcinogenesis of cholangioncarcinoma.
