| Objectives: To investigate the effects of alprostadil and noradrenaline and anisodamine on model of rat septic shock,by comparing induced nitricoxide synthase(iNOS) level in nephridial tissue and activity of mitochondrial respiratory chain complex I in nephridial tissue and cystatin-c level in serum, to evaluate the roles of all those medicines on renal microcirculation and glomerular filtration rate (GFR).Methods:Fifty male S-D rats weight 250-280g were randomly divided into five groups, each group includes ten rats. control group(groupA),septic shock group (groupB), treatment group with alprostadil (groupC),treatment group with noradrenaline(groupD),treatment group with anisodamine (group E),Each group was divided into 1h and 3h subgroups (n=5 in each subgroup). Septic shock model was made by injecting lipopolysaccharide(LPS) via left femoral vein continuedly.Group A was given normal sodium also via left femoral vein continuedly; a tube was establish in left carotid artery in each rat,blood pressure can be measured by using those tubes. Incision of trachea was also employed in all rats.In group B,LPS was injected via left femoral vein by the rate of 10mg/kg.Alprostadil was injected via left femoral vein by the rate of 50ug/kg.h in group C.Noradrenaline was injected by the rate of 0.1mg/kg.h in group D. Anisodamine was injected via left femoral vein by the rate of 0.1mg/kg.h in group E. group A and group B were injected equal volume by equal rate.All rats were killed after 1h and 3h.serum was taken out and used to measure cystatin-c by means of ELISA method.Renal was homogenated after velamen and renal pedicle were removed,then homogenation was used to measure iNOS and activity of mitochondrial respiratory chain complex I.Results:1 Cystatin-c level in serum in group B and group D were heightened gradually with the time prolonged, the difference between group B and group D was quiet,cystatin-c level in group Cand groupE in 1h and 3h also heightened, but compared with those in group B and group D, cystatin-c level in group C and groupE were lower, difference was significant.2 Activity of mitochondrial respiratory chain complex I in nephridial tissue in group B and group D were degraded gradually in 1h and 3h, which were strengthened in group C and group E, difference was significant.3 iNOS in nephridial tissue in group B and group D were heightened, which was also heightened in 1h in group C and group E, but degraded in 3h, difference was significant. compared with group D and group B, iNOS in group C and group E were lower, difference was significant. Conclusions:1 A septic shock model can be made successfully by injecting LPS via vein.2 Anisodamine and alprostadil all can improve renal function and activity of mitochondrial respiratory chain complex I in septic shock, maybe because of microcirculation was improved.3 Anisodamine and alprostadil can lower iNOS level in nephridial tissue, maybe owing to rival the effects of ET, injure of microcirculation can be lessened.4 there is seemly no obvious effects of norepinephrine on improving activity of mitochondrial respiratory chain complex I and renal function, it also seems no obvious effects on lessening iNOS level in nephridial tissue.maybe in early phase of shock, ET plays an important role, it contracts blood vessel and norepinephrine can not improve organ perfusion. |
