| Objective: To investigate the experimental effect on rat renal calculus treated by Bushenhuashi Decoction and to analyze the possible therapeutic mechanism.The model of kidney stone was made by imbibing the water which included 1% glycol and 1% ammonium chloride.And some indexes such as content of oxalic acid ,calcium in kidney,the degree of kidney calculi ,the rat of stone of fomation,the content of serum mg ca BUN Cr and renal pathomorphology were observed, which would provid experimental data and evidence for clinical application of Bushenhuashi Decoction.Method:The experiment were performed on 40 healthy male SD rats,weighed 200~220g.The rats were fed with common feedstuff for 1 weeks and then randomly divided into four groups:Normal group,Model group,Bushenhuashi Decoction group (BD group) and Paishi Granule group (PG group),free eating and drinking(10 rats each group).Other groups except the Normal one were subjected to modeling.The Model group was made by imbibing the water which included 1% glycol and 1% ammonium chloride for 5weeks.The drug was given to the rats in other all groups except the Normal one after one week of the beginning of the modeling,then the administration was continued.Normal group and model group were poured equal dose distilled water.Bushenhuashi Decoction group were poured gastrically perfused with the traditional chinese medicine concentrated decoction 2.89g/ml one time per day.Paishi Granule group were poured paishi granule 2ml which concentration was 0.54g/ml.The rats were sacrificed at the fourth week after medication for getting blood throuth carotid artery cannula.The serum mg ca BUN and Cr were assayed with the automaticbiochemistry.The pathomorphological change of nephridial tissue was observed through light microscope, electron microscope,and also the level of mesoxalic and calcium was detected.Result: (1) The effect on content of mesoxalic and calcium in nephridial tissue of rats:The content of calcium and oxalic in renal of Model group was higher than Normal group significantly(P<0.01);The content of calcium and oxalic in renal of Bushenhuashi Decoction group descended significantly compared with Model group (P<0.01);The content of calcium and oxalic in renal of Paishi Granule group descended significantly compared with Model group (P<0.01);However,the two kinds of drug decrease the calcium and oxalic content in renal , Bushenhuashi Decoction group is much better than Paishi Granule group(P<0.01).(2) The effect on serum biochemical index in rats:the level of BUN and Scr in blood serum of Model group was higher than Normal group significantly (P<0.01).BUN, Scr and ca of Bushenhuashi Decoction groups descended significantly compared with Model group (P<0.01).The content of mg was higher than Model group (P<0.01).BUN, Scr and of Bushenhuashi Decoction groups descended significantly compared with Paishi Granule group (P<0.01),the content of mg was higher than Paishi Granule group (P<0.05).(3) The extent of rat kidney stones and rate of formation each group was compared:Crystallization were not observed in Normal group,a large number of CaOx crystals were observed in Model group,the Crystallization distributed in bulk in most renal,Crystalline powder are not connected,some individual Crystallization connected inbuted,the rate of stone formation was 100%;only a few CaOx crystals were observed in Bushenhuashi Decoction group and Paishi Granule group,the rate of stone formation was 37.5% and 50%.Bushenhuashi Decoction Was effective in preventing and treating Urolithiasis of rats induced by glycol,There was a significant difference between Bushenhuashi Decoction group and Model group (P<0.005),the rate of stone formation and the degree of stone formation of the Bushenhuashi Decoction group is lower than Paishi Granule group,but there was no significant difference between them.(4) Pathomorphological changes of Nephridial tissue.Renal tubular atrophy:From light microscope view we could see significant tubular dilatation and edema of the epithelial cells were observed in the model group,a large number of hybridization CaOx crystals and necrosis epithelial cells were observed in renal tubular and a large number of inflammatory corpuscle infiltrated.From electronic microscope view we could see amount of microvilli on kidney proximal tubule surface decreased significantly,the microvilli lined up in disorder,some cristae and membrance disappeared or was portmanteau in mitochondria, the rough endoplasmic extended, the doff particle phenomenon was significantly and cells of connection are broaden. In the Bushenhuashi Decoction group, the electronic and light microscope observation result showed that: tubular dilatation and edema of the epithelial cells were obviously alleviated, which had remarkable difference compared with the model group,but only a few CaOx crystals and a limited distribution of CaOx crystal were detected.A few of chronic inflammatory corpuscle infiltrated, there were counts of numerous microvilli on the free surface of kidney proximal tubule.The microvilli lined up in order and the phenomenon of doff particle was obviously alleviated. A small part cristae and membrance disappeared or was portmanteau in mitochondria, which had obviously alleviate compared with the model group, significantly difference compared with Paishi Granule group.Conclusion:The results demonstrated that:Bushenhuashi Decoction has the superiority of adjusting the human body as a whole. It can prevent kidney stone,relieve pain symptons and increase urine column drain stone. It can resolve stone and nourish shen to protect renal tuble cell working normally, prevent cell dying.The serum biochemical index suggest that Bushenhuashi Decoction can help the rats with Nephroplithiasis nephritis to decrease the rat's BUN and Cr, the function of glomerular filtration was improved,to protect renal function and inhibit renal failure.Its whole effect is superior to the Paishi Granule.All these illustrate that Bushenhuashi Decoction can prevent the Nephrolithasis and complications.The concrete mechanism is yet to he confirmed. |
