Study On The Isolation, Purification And Anti-tumor Activities Of Polysaccharides From Scurrula Parasitica L | | Posted on:2009-07-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y L Fan | Full Text:PDF | | GTID:2144360245485130 | Subject:Developmental Biology | | Abstract/Summary: | PDF Full Text Request | | Scurrula parasitica L.is one of the most common parasite scurrula plants in the austral area of our country.In our early empirical studies;we found the total flavonoids in Scurrula parasitica L.had favourable anti-tumor effects in vivo and vitro,Viscum album L.plants,sibling plants of Scurrula parasitica L.,which is an orthodox anti-tumor plant amedica in Europe.The extract of Viscum album L.had cytotoxic and immunoregulatory effectiveness and was admoved as adjunctive therapy chemotherapeutics for clinical anti-cancer curatio.We have maded much research as follow:the isolation,purification of polysaccharides from Scurrula parasitica L.,its physico-chemical property,its anti-tumor effectiveness in vivo,its combination chemotherapy effectiveness with Cytoxan and the initial mechanism of anti-tumor.The optimum extraction and isolation conditions for polysaccharides from Scurrula parasitica L.were established according to one-factor and orthogonal experimentation. Two main fractions of polysaccharides from Scurrula parasitica L.were obtained after eluting from DEAE-SephadexA-25 and Sephadex G-100 column chromatography.Thc physicochemical characterization and part of structure characteristics of polysaccharides from Scurrula parasitica L.were analyzed by ultraviolet spectra and infrared spectrum.The anti-tumor effects of polysaccharides from Scurrula parasitica L.were detected by establishing the mice-transplanted tumor models with injecting the sarcoma 180 ascites tumor at the right oxter of mice,and the effect on immune organ weight were examined by spleen and thymus index.Polysaccharides from Scurrula parasitica L.was given with the Cytoxan to detect the inhibition effect of the the sarcoma 180 solid tumor. In the results,polysaccharides from Scurrula parasitica L.could significantly increase the inhibition effect of the the sarcoma 180 solid tumor,spleen index,thymus index and the anti-tumor effect of Cytoxan at the dose of 100 mg/kg.d.Polysaccharides from Scurrula parasitica L.also could markedly improve the immune inhibition of Cytoxan.The expression of Ki-67,Cyclin D1,Bax and Bcl-2 protein in the sarcoma 180 solid tumor were detected by immunohistochemical techniques to approach the anti-tumor mechanism of polysaccharides from Scurrula parasitica L..The integrated optical density was measured by image analysis system.In the results,one antineoplastic mechanism of polysaccharides from Scurrula parasitica L.may accelerate the apoptosis of tumor cell and inhibit tumor tissue growth by down-regulating the expression of Ki-67, CyclinD1 and Bcl-2 protein and up-regulating the expression of Bax protein. | | Keywords/Search Tags: | Scurrula parasitica L., polysaccharides, orthogonal experiment, the sarcoma 180, Cytoxan, combination chemotherapy, immunohistochemistry | PDF Full Text Request | Related items |
