| Objective: Copy the model of the chronic arsenic poisoning rats and then investigate the effect of CCK on sensory disturbance induced by the chronic arsenic poisoned so that to supply the proof for the clinical therapy and search for the effective drug for the chronic arsenic poison. Methods:1,To set up the method of DRG culture and identificate the neurons use NSE staining.2,After successful copied the model of chronic arsenic poisoning rats,pretreated with CCK and culturing the DRG neurons of the E16-18 d fetus rat,to observe the effects of CCK on the survival time of the DRG neurons .3,To detect the expression of CCK on the DRG neurons by IHC method and detect the positive cell photodensity value of CCK expression of every group DRG neurons.4,To detect the AP and CV in the sciatic nerve with electrical physiology method and to investigate the effect of CCK on them.Results: 1,Cultured the DRG neurons and identificated the positive neuronal specificity staining by NSE successful .2,After pretreated with CCK,the survival time of DRG neurons in the groups of LD and MD were prolonged from 154±3 h to 186±4 h (P < 0.01),79±4 d to 114±2 h(P < 0.05) respectly. But After pretreat with CCK,the DRG neurons'survivial time in the group of HD has not evident change.3,The expression of CCK on the DRG neurons was enhance in every group compared with the control group (P < 0.05). The positive cell photodensity value of CCK expression of every group DRG neurons were 0.163±0.002,0.196±0.003,0.274±0.011,0.201±0.004 respectly. The positive cell photodensity value of CCK expression in MD group is the highest among the total groups.4,The amplitude of AP and CV of chronic arsenic poisoning rats display a descent tendency company with the increasing arsenic poisoning degree(P < 0. 01). In LD and MD groups, they are different when used CCK or not (P < 0. 01). But in HD group, they are not evident change whether in used CCK or not (P > 0. 05).Conclusions: 1,Comparing with the control group, the survival time of DRG neurons are shortened in chronic arsenic poisoning fetus rats.2,CCK play a protect effect on the model of the cultured chronic arsenic poisoned DRG neurons , and it can prolong the survival time of DRG neurons in LD and MD groups.3,The expression of CCK on the chronic arsenic poisoned DRG neurons was enhanced , especially in the Small neuron. These changes confirmed that CCK play a vital role in sensory disturbance induced by the chronic arsenic poisoned.4,The chronic arsenic poisoning can effects the AP and CV in the sciatic nerve of rat. CCK can partly recover the amplitude of AP and CV.Above the researches may supply the theory proof for the sensory disturbance induced by the chronic arsenic poisoned for clinical therapy and CCK may be a effective drug for the chronic arsenic poison. All the results have significance whether in theory or clinic practices. |
