Objective: To synthesis Magnesium TanshinoneⅡA Sulfonate and establish its quality control methods; To investigate the protective effects and its protective mechanism on acute myocardial infarction in rats.Methods: The Magnesium TanshinoneⅡA Sulfonate was synthesised by ion-exchange methods and its power of hydrogen, pyretogen and abnormal toxicity were test. It was determinded by high performance liqiud chromatography method; Precision, stablity and repeatability of Magnesium TanshinoneⅡA Sulfonate were investigated respectively.The acute myocardial infarction model in rats were performed by the ligation of the left coronary artery and the successful models were randomly assigned to control group (5% glucose solutions), Sodium TanshinoneⅡA Sulfonate group(5mg·kg-1),and Magnesium TanshinoneⅡA Sulfonate group(1,5,15mg·kg-1). The corresponding dosages medicine were given to that rats 5 minutes after ligationing through the vena caudalis; The myocardial infarct size, changes of lactic acid dehydrogenase and creatine phosphokinase activity in serum, superoxide dismutase activity and malondiadehyde contents in myocardium were simultaneously monitered 6 hours later.Results: The yield and purity of the product were 93.62% and 93.39%, respectively. There was a good linear relationship within range of 0.6531~10.4500μg·ml-1 (r=0.9996). The average recoveries were 100.15%with corresponding RSD of 1.06%. The results of precision, stablity and repeatability test were 0.97%, 1.29% and 1.75%, corresponding test requirement. Comparing with control group it was found that Magnesium TanshinoneⅡA Sulfonate decreased the lactic acid dehydrogenase and creatine phosphokinase activity in serum ,reduced myocardial infarct size, and improved the activity of superoxide dismutase and decreased the malondiadehyde contents in myocardium respectively.Conclusions: The preparation metheod is simple and feasible. The determination method is sensitive and accurate, which can be used for the quality control of Magnesium TanshinoneⅡA Sulfonate. MTS had protective effects on the acute myocardial infarction by the ligation in rats, the mechanism of the protective effect may be related to its role against oxygen free radical.
