| Objective: to study the disturbing effect of the common chemotherapeutics such as DDP,ADM and 5Fu on the expression of SNCG of the MDA-MB231 breast cancer cells.Methods: EC50(Concentration for 50% of maximum effect I.e. concentration inducing death of 50% cell)for MDA-MB231 breast cancer cell of three chemotherapeutics , DDP,ADM and 5Fu ,were tested with MTT. MDA-MB231 breast cancer cells were treated with 1/2EC50 forementioned chemotherapeutics respectively, samples were taken after 72h. RT-PCR and immunohistochemical method were used to detect the expression of the SNCG mRNA and protein in MDA-MB231 cells in the study group treated with the drugs above and the negative control group. The expression level of the SNCG mRNA and protein of every group were analysis with the Quantity One software and CM-2000B system respectively.Result: The expression of the SNCG mRNA and protein of the groups treated with ADM and 5-Fu were lower than the negative control group(P<0.05), with statistically significant difference. While there was not difference between the group treated with DDP and the control group.Conclusion: The expression of SNCG of the MDA-MB231 breast cancer cells can be inhibited by ADM and 5Fu, while DDP has no effect on its expression. |
