| PART ONERESEARCH ON BLOOD-BRAIN BARRIER IN TRACRANICAL TUMORS OF CHILDRENObjective: To investigate the mechanism of the multidrug resistance in intracranial tumors, by detecting the expression of the multidrug resistance protein in children's intracranial tumors and normal brain tissue.Methods: The expression intensity and disposition of P-glycoprotein (P-gp), multidrug resistance-associated protein (MRP), Lung resistance-related protein (LRP), Topoisomerase II (TopoII) and Glutathione-S-transferase-Ï€(GST-Ï€) in 30 patients and 5 normal brain tissue were detected by SP immunohistochemistry.Results: The high expression of P-gp was observed in capillary walls and tissue around vessels, while the low expression was observed in the tumor cells. The expression of other multidrug resistance protein couldn't be observed in vessel walls.Conclusions: The chemotherapeutics drugs were interrupted in the capillary endothelium cell and the end-feet of neuroepithelial cells in the blood-brain barrier by P-gp, which weakened the chemotherapeutics effect. TopoII was related to the mechanism of the multidrug resistance in intracranial tumors. PART TWOAPPLICATION OF HISTOCHEMISTRY IN THE DIAGNOSIS AND DIFFERENTIAL DIAGNOSIS OF HIRSCHSPRUNG DISEASEObjective: To research the application of histochemistry in diagnosis and differential diagnosis of Hirschsprung's disease (HD) and Hirschsprung's allied disease (HAD), by observing the expression of various antibodies and the acetylcholine esterase (AchE) staining.Methods: Bcl-2, calretinin (CR), neuron specific enolase( NSE) , S-100 proteinum and catepsin D( CAD ) were used to mark the intestinal wall of the distension segment, the transmigration segment, and the spasm segment which were stained by the osmium black formation AchE staining method and the rapid AchE staining method meanwhile, after the radical correction of HD .Results: Clearer result can be get in the osmium black formation AchE staining method, while less time was taken in the rapid AchE staining method. Bcl-2 was only positive in the degenerative and immature neuro ganglion cell, CR was negative in the neuroplexus in the intestinal with degenerative neuro ganglion cell. NSE,S-100 and CAD were complementary to each other to observe the neuroplexus. Conclusions: The osmium black formation AchE staining method can be used for diagnosis after the HD operation, while the rapid formation is suitable for diagnosis on operation. And the specific expression of Bc1-2 and CR took an essential role in the diagnosis of HAD and newborn HD. |
