| Objective : To study the expression of discoidin domain receptor 1 levels in esophageal squamous cell carcinoma (ESCC), the correlation between expression of DDR1 protein in ESCC, and relationship with clinicopathologic characters,such as depth of invasion, histologic grade, lymph node metastasis and to investigate it's role in tumorigenesis as well as progression in ESCC and their clinical significant.Method : The expression of DDR1 protein was detected in 40 cases of esophageal squamous cell carcinoma tissues,the para-cancer tissues and matched normal esophageal tissues by immunohistochemistry and western-blot , respectively , and their relation to clinical pathologic characteristics of ESCC was analyzed.Result :1. DDR1 protein expression was observed in the cell membran, The overexpression of DDR1 protein determined by western blot analysis was similar to that by immunohistochemical analysis, demonstrating that the expression of DDR1 in cancer was significantly higher than in the corresponding non-cancerous tissues and normal tissues . 2.The expression rate of DDR1 in ESCC (25/40,62.5%) was Significantly higher than that in the para-cancer tissues(17/40,32.5%) and normal esophageal tissues(8/40,20.0%)(χ2=14.035,P <0.005).3. The expression of DDR1 in ESCC (1.201±0.348)were significantly higher than those in para-cancer tissues (0.640±0.323)and normal esophageal tissues (0.551±0.413) by western blot analysis. DDR1 protein was expressed significantly higher in moderate/poor differentiated tumors than that in well-differentiated (t=3.7811, P<0.01), The expression of DDR1 in the group of lymph node metastasis were significantly higher than those without lymph node metastasis(t=6.9517, P<0.01). The positive expression rates in samples of different tumor infiltrating depth of esophageal SCC was different(P<0.05).The positive expression rate and relative content of DDR1 in ESCC were correlated with degrees of differentiation,lymph node metastasis and infiltrative depth of carcinoma (t=3.7171, P<0.01). There were no significant in different in sex,age and the length of ESCC(P >0.05).Conclusion: The expressions of DDR1 protein increased gradually from normal tissues, para-cancer tissues to carcinoma tissues. It was correlated with carcinoma differentiation,nodal metastasis and the depth of invasion. Indicating DDR1 protein was related to ESCC, development and invasion, metastasis,DDR1 protein maybe act as one of biological indicators of the invasion and metastatic potential in ESCC. |
