| Hepatocellular carcinoma(HCC) is one of most common malignant tumour harming our health. At present, two basic principles have been generally acknowledged on the treatment of HCC. Firstly, radical excision is still the most effective means in promoting the long term survival. Secondly, we cannot achieve best effect by single treatment but combined therapy. Hepatic artery chemotherapy embolism is an important therapy in combined therapy. Pingyangmycin ,because of the effect of both anti tumour and blood vessel embolism, has its superiority in the treatment of HCC,which is a kind of tumour rich in blood vessel. Thalidomide,which is also an immunoregulant,can inhibit angiogenesis. Combining pingyangmycin with thalidomide in treating HCC may bring good results.Purpose: to observe the effect of the combination of pingyangmycin and thalidomide in the treatment of murine H22 transplanted liver carcinoma , the tumour MVD and the expression of VEGF,the correlation betwwen MVD and VEGF and to investigate the antitumour mechanism involved.Method: 1,H22 hepatocellular carcinoma cells were cultured in vitro. 2,Murine H22 transplanted tumour model was established subcutaneouly, the mice bearing tumour were divided into 6 groups, Group A: control group; Group B: lipiodol group; Group C: thalidomide group; Group D: pingyangmycin group; Group E: thalidomide+pingyangmycin; Group F:thalidomide+pingyangmyci+lipidol.Thalidomide was administrated intragastricly, lipiodol and pingyangmycin were injected intratumorally. The tumour's diameters were measured every other day from the 7th day after the mice were innoculated, and all the mice were sacrificed on the 14th day. The stripped tumour were weighed. the tumors were pathologically sectioned, HE stained after fixed by 10% formalin. Tumour MVD were detected by immunohistochemistry staining with CD34 antibody and the expression of VEGF were observed by immunohistochemistry staining with rat VEGF multiclone antibody. Results: 1,Tumour of group C,D,E,F grew slower than group A. there was no obvious difference between group B and group A .tumour inhibiting rate of group B,C,D,E,F was respectively 6.0%,56.80%,57.40% 41.77% 38.77%, there existed obvious difference of tumour weight in group C,D,E,F compared with group A, but there was no statistical significanc(eP﹥0.05)between group B and group A. MVD of A,B,C,D,E,F was separately 40.70±3.45,39.58±2.48,21.96±3.38,23.39±3.04 24.45±2.91,25.55±2.36, and the evaluating score of the expression of VEGF was separately 6.00±0.00,5.92±0.20 ,0.75±0.61,1.31±1.03,1.67±1.03,2.17±0.98。Conclusion: The combination of pingyangmycin and thalidomide can inhibit tumour growth in murine H22 subcutaneouly transplanted hepatocellular carcinoma. The mechanism of antitumour may due to inhibiting Angiogenesis. The expression of VEGF was positively correlated with MVD... |
