1-Deoxynojirimycin(DNJ) from mulberry leaves is one of the useful compositions which can decrease blood sugar , and has various pharmacological activities, such as anti-hyperglycemia,anti-cancer,antivirius and so on. Therefore, it has great value to extract,separate,purify DNJ from natural mulberry leaves and establish the method of content determination for preparing new drugs curing diabetes and developing the relative products of mulberry leaves. The chemical study on DNJ is the main contents of this paper.Firstly, it is the first time to extract DNJ from mulberry leaves using chemical reaction and column chromatography. DNJ and its structural analogue 1,2-dideoxynojirimycin (Fagomine) were isolated and purified from mulberry leaves by the method of water extracting-alcohol precipitating and various chromatographic methods, such as silica gel column chromatography, ico exchange column chromatography and gel column chromatography . The purity of DNJ is above 98% according to area normalization. Based on experiments, the method of content determination:HPLC-ELSD was established which was easily to operate, accurate and repeatable comparing methods available.And it can be applied to the content determination of DNJ from mulberry leaves and other relative products.Secondly, it was the first time to compare the DNJ contents in mulberry leaves from different areas by the method of HPLC-ELSD and compare the differences between mulberry leaves and excrementum bombycis in the same area. The results showed that different areas revealed different contents. Among them, Zhejiang-DNJ content was higher than other areas followed by Chongqing,Shanxi and Hebei. Maybe the different climate leads to the main differences, but concerning the same region , the DNJ content in excrementum bombycis was distinctly lower than mulberry leaves.At last, the derivative of DNJ was synthesized , and the product N-Ethyl-DNJ was obtained and identified by MS and NMR datas.
