| Objective: Toll-like receptor 4 is a member of the toll-like receptor family, It is a typeâ… transmembrane receptor, after recognizing exogenous or endogenous ligands , It would activating the intracellular signal transduction pathway, promoting special intranuclear gene expression, and secreting Th1 cytokines such as IL-1,IL-6,IL-12,TNF-a,IFN-γand so on. Recent research shows that Th2 cytokines is prevail in normal pregnancy, and shift of Th2/Th1 cytokines, contribute to immune imbalance and spontaneous abortion. In view of this background and condition, we put forward a hypothetical mechanism named TLR4-Thl cytokines-spontaneous abortion mechanism. The expression of toll-like receptor 4 was observed in decidua and chorionic villi tissues in patients with early spontaneous abortion, and then analysising the mechanism of spontaneous abortion, accordingly, it would supply new breakthrough of clinical diagnosis and treatment on spontaneous abortion.Methods: 30 women suffering spontaneous abortion and 20 women with normal pregnency were randomly selected accordance with inclusion criteria . The sample is obtained during artificial abortion, then it is bathed by normal sodium, fixed by 10% formalin. Immunocytochemistry was performed to detect the expression of TLR4 in human villus and decidua . All data were analyzed by SPSS 12. 0 software, rank-sum test is used for data. P<0.05 is considered as significant difference.Result: 1,The expression and localization of TLR4 in human villus and deciduas: Either in normal or spontaneous abortion women , TLR4 can express in villus and deciduas tissue, The immunohistochemistry indicated that the coloration of TLR4 is in the endochylema and membrane of the cell , the expression of TLR4 was mainly located in the syncytiotrophoblast in human villus, while in deciduas was mainly in gland, especially in epithelial cells.2,The expression intensity of TLR4 between the two groups: compared with the normal pregnant group , the expression intensity of TLR4 in villus is higher in spontaneous abortion group, the difference is significant (P<0. 01) ;and, the expression of TLR4 in deciduas is also higher. The difference is significant (P<0. 01). The expression intensity of TLR4 in chorionic villi was parallel to and was positively correlated with that of in decidua.Conclusion: 1,TLR4 is expressed in villus and deciduas tissue of normal pregnant women.2,The expression of TLR4 in the tissue of villus and deciduas is higher in spontaneous abortion group than the normal pregnant group, indicated that Toll-like receptor 4 may be one of the mechanisms lead to abortion.3,The pathogeny of spontaneous abortion is complex, the hypothetical mechanism we put forward may be one of these. |
