【Background】Pathological scars, including hypertrophic scar and keloid, can be seen quite often in clinic work. It is often accompanied with dyskinesia or dysfunction of limbs. Because the exact pathological mechanism underlying the two lesions is unknown, there are a great number of treatment modalities but none can treat the pathological scar successfully. Curcumin, a nature-pigmented extracted from the perennial herb of Curcuma species, is an active principle of turmeric. It is reported that curcumin might be a potential antifibrotic drug. However, it has not been reported that whether curcumin can inhibit aberrance fibroblast and be used to treat pathological scars.【Purposes】1. To study the effects of different concentration of curcumin on the proliferation and the function of synthesis collagen of hypertrophic scar fibroblasts in vitro. To investigate the mechanism of curcumin in treatment of hypertrophic scar.2. The KFB were treated with various concentrations of curcumin. To investigate the effects of curcumin on the proliferation and the function of synthesis collagen in keloid-derived fibroblast. To investigate the mechanism of the effect of curcumin on KFB.【Contents】1. The biological effects of curcumin on hypertrophic scar fibroblasts in vitro; 2. The biological effects of curcumin on keloid fibroblasts in vitro.【Results】1. The proliferation of the HSFB in vitro can be inhibited significantly by curcumin. The apoptosis of the HSFB in vitro can be accelerated significantly by curcumin. Meanwhile, curcumin can inhibit the expression of the amount of Bcl-2, which is regarded as one of the functional way of curcumin;2. Curcumin can obviously inhibit the expression of the amount of typeⅠandⅢprocollagen mRNA of fibroblasts derived from hypertrophic scar;3. Curcumin can obviously inhibit the proliferation and the synthesis collagen in keloid-derived fibroblast;4. Curcumin can obviously inhibit the expression of the amount of typeⅠandⅢprocollagen mRNA of fibroblasts derived from keloid. Meanwhile, curcumin can inhibit the expression of the amount of CTGF, which is regarded as one of the functional way of curcumin.【Conclusions】1. The HSFB in vitro can be inhibited by curcumin. The apoptosis of the HSFB in vitro can be accelerated significantly by curcumin. The mechanism of curcumin to induce HSFB apoptosis may be associated with the down-regulating expressions of bcl-2 on HSFB;2. The KFB in vitro can be inhibited by curcumin. Maybe its mechanism is inhibiting the expression of the connective tissue growth factor.
