Objective: To study pharmacokinetics of levofloxacin and the effect of Chinese herb Achyranthes bidentata on pharmacokinetics of levofloxacin in pathological model of rats with prostatitis.Methods: 78 pathological model of rats with prostatitis were allocated into two groups of 39 rats each at random.The rats in groupⅠwere orally given with single dose of levofloxacin at 80mg/kg·b·w,the rats in groupⅡwere orally given with single dose of levofloxacin at 80mg/ kg·b·w and Achyranthes bidentata at 2g/kg·b·w simultaneously. The concentrations of Levofloxacin in plasma and tissue were determined by high performance liquid chromatography with a limit of detection of 0.001μg/L and 0.001μg/g, respectively.The pharmacokinetic parameters were calculated by using DAS2.0.1 pharmacokinetics software.Results: Concentration-time data of all groups were best described by a two-compartment open model with 1st order absorption(weight=1/cc).The main Pharmacokineticsparameters and calculat ed-equations in rats were as follows,respectively.Group I :T1/2Ka=(45.294±2.07)min,T1/2α=(63.963±15.258)min,T1/2β=(280.535±214.601)min,Tmax=(50±17.321)min,Cmax=(14.905±3.143)mg/L,AUC (0→2160)=(2561.219±430.592)(mg·min·L-1),Ka=(0.015±0.001)(1/min),MRT0→2160=(196.865±50.65 6)mm,C=66.697·e-0.011t+6.031·e-0.005t-72.728·e-0.015t.Group II:T1/2Ka=(21.077±4.554)min,T1/2α=(94.6 80±47.380)min,T1/2β=(637.802±624.798)min,Tmax=(60.000±0.000)min,Cmax=(12.771±1.620)mg/L, AUC0→2160=(2796.476±450.485)(mg·min·L-1),Ka=(0.034±0.007)(1/min),MRT0→2160=(257.825±6 1.315)min,C=23.236·e-0.007t+2.442·e-0.001t-25.678·e-0.034t.Conclusions:①Levofloxacin in in pathological model of rats with prostatitis was quickly absorbedwith high concentration in plasma,distributed widespread,and eliminated slowly.②Achyranthes bidentata could influence the pharmacokinetic parameters of levofloxacin in rats with prostatitis significantly: decreased the speed of absorption,but enhanced the degree of absorption,accelerated the distribution from blood to tissues,retarded the speed of elimination, increased the time of drug action.③Achyranthes bidentata leading action could notably influence the distribution of levofloxacin in rats with prostatitis.Increased the drug concentration of levofloxacin in parastata tissue,increased the concentration of liver tissue, increased the drug concentration of kindey tissue.④Achyranthes bidentata could notably influence the distribution of Levofloxacin in in rats with prostatitis.
