| Portulaca oleracea L.belongs to the genus Portulaca(Portulacaceae),which is distributed in most areas all over the world.It has long been used as traditional Chinese medicine in China.Except antibiosis,blood-fat,antiinflammatory,analgesia,facilitate intention effects,the herb also showed anti-hypoxia and anti-aging effects.In the pharmacological study,the extract of Portulaca oleracea L.showed strong anti-hypoxia activity.For further exploitation of this plant,the investigation of chemical constituents and quality analysis of Portulaca oleracea L.were carried out.Our research results are described as follows:1.the chemical components10 components were isolated from the herb of Portulaca oleracea L..The structures of 8 compounds were elucidated,including 2 new components.By chemical and spectroscopic methods(1H-NMR,13C-NMR,HHMQ,HMBC,1H-1HCOESY,NOESY,EI-MS,IR and UV ),2 new compounds were identified as 3-O-β-D-glucopyranosyl(1-4)-β-D-xylopyranosyl-(2α,3α)-2,3, 23-trihydroxyolean-12-en-28,30-dioicacid-30-methylester(1),3-O-β-D-xylopyranosyl -(2α,3α)-2,3,23,30-tetrahydroxyolean-12-en-28-oic acid(2),along with 6 known compounds:β-sitosterol(3), 1-triacontanol(4),4a-methyl-3β-OH-friendelan(5),β-daucosterol(6)and 4-hydro xy phenethylamine(8).By GC-MS method,chemical constituents with low polarity in extract of Portulaca oleracea L.were investigated.51 chromatographical peaks were separated and 44 chemical constituents were identified including palmitic acid 17.06%,ethyl linoleate 16.04%,palmitic acid,ethyl ester 14.47%,α-linolenic acid 12.72%,linoleic acid 7.66%,whose content were determined with peak area normalization method.These chemical constituents were mainly saturated fatty acid ester,unsaturated fatty acid ester and alkanes.2.the quality analysisSet up the quality criteria of both Portulaca oleracea L.herb and extract.Qualitative and quantitative analysis were carried out withα-linolenic acid as the authentic.Thin layer chromatography showed thatα-linolenic acid can be detected both in Portulaca oleracea L. herb and extract.3.the quality evaluationDetermination of 25 samples of Portulaca oleracea L.indicated that the content of unsaturated fatty acid and flavone changed depending on the locations.4.the preparation technologyTo optimize the preparation technology of extract of Portulaca oleracea L.with extraction yield ofα-linolenic acid,the content ofα-linolenic acid and the yield of extract as criterias,whose contents were determined by HPLC,the best preparation process conditions were optimized in L9(34)orthogonal test,combining with the pharmacological results.The preparation technology conditions were verified consistent,convenient and practical. |
