The Relationship Of CD4~+CD25~+ Regulatory T Cells And Controlling Gene FOXP3 With Graft-Versus-Host Disease

Objective: To study the expression levels of CD4~+CD25~+Tregs and FOXP3 in graft-versus-host disease (GVHD) patients with the Allogenetic peripheral blood stem cell transplantation (Allo-PBSCT) ,and investigate the relationship of CD4~+CD25~+Tregs and FOXP3 with GVHD.Methods: The expression levels of CD4~+CD25~+Tregs and FOXP3 in 42 patients with allo-PBSCT were detected by the immunofluorescence and flow cytometry.The expression levels of FOXP3 were measured by the real-time quantitative reverse transcription polymerase chain reaction(real-time RT-PCR).The specificitis of products were identified by The Melt Curve and The agarose gel electrophoresis.Reasults: 1,In the aGVHD group, compared with the Non-aGVHD group, there was no significant difference of the levels of CD4~+CD25~+Tregs in +30d,+60d,+90d (P > 0.05) ,but was the obvious decrease of expression levels of FOXP3 showed in +60d,+90d (+60d: 1.31±0.12 VS 2.41±0.15, P<0.05; +90d: 2.52±0.80 VS 5.42±0.07, P<0.01) .2,In the cGVHD group, the levels of CD4~+CD25~+Tregs were notobviously different in +60d,+90d compared with the Non-cGVHD group(P > 0.05) ,but ware increased in +180d (12.37±0.31 VS 10.35±0.52,P<0.05). The expression levels of FOXP3 in the cGVHD group and the Non- cGVHD group were not significantly different in+60d,+90d (P > 0.05) , but revealed fewer in the cGVHD group (1.27±0.10 VS 6.58±0.09, P< 0.01) .3,The levels of CD4~+CD25~+Tregs in the aGVHD group were positively correlated with those of FOXP3 (Pearson:0.926, P=0.000) ,and were not correlative in the cGVHD group (Pearson:0.315, P=0.074) .4,The expression levels of FOXP3 in theⅡ+Ⅲ°aGVHD group were significantly lower than those in theⅠ°aGVHD group (P<0.05), while also significantly lower than those in the Non-GVHD and health persons (P<0.01) .Conclusion: 1,The levels of CD4~+CD25~+Tregs in the aGVHD group and the Non-aGVHD group were not significant deviation (P > 0.05) , The expression levels of FOXP3 in the aGVHD group were lower than those of in the Non-aGVHD group (P<0.05) .2,The levels of CD4~+CD25~+Tregs in the cGVHD group were higher than those of CD4~+CD25~+Tregs in the Non-cGVHD group, The expression levels of FOXP3 in the cGVHD group were lower than those in the NoncGVHD group (P<0.05) .
