The Experimental Study About The Influence Of B(a)P To The Expression Of The Aurora-A Protein In The Xuanwei Lung Cancer-05

Objective:To study the expression of the Aurora-A protein in the adenocarcinoma of lung of Xuanwei Lung Cancer-05 and the influence of Benzo(a)ypyrene to the expression of the Aurora-A protein in the cells of adenocarcinoma of Xuanwei Lung Cancer-05.Method:1,Type theâ…¡alveolar epithelial cells,lung cancer cells(A549)and Xuanwei Lung Cancer-05(XWLC-05)were cultivanted and spreaded to the 20th generation.Then the expression of Aurora-A Protein in the three kinds of cells was detected by flow cytometry.2,The inhibition rate of the different concentrations of Benzo(a)ypyrene to the XWLC-05 was determined through MTT method.According to it,the most suitable action concentration of Benzo(a)ypyrene to the growth of XWLC-05 was choosed.3,XWLC-05 cells were separated into two sets:A and B.Every set included six kinds of action concentration groups of Benzo(a)ypyrene,that was from N₁ to N₆.The A set acted 24 hours,while B set acted 48 hours.Then the changes of the Aurora-A protein's expression in the XWLC-05 of every group were detected by flow cytometry.Results:1,The expression of Aurora-A protein in XWLC-05 cells was the strongest,that was the second in the A549.And it was the weakest in human typeâ…¡ alveolar epithelial cells.2,The inhibition rate of XWLC-05 cells is may less than 15%,when the concentration of Benzo(a)ypyrene less than 200umol/L.So six kinds of concentration of Benzo(a)ypyrene were chosed to deal with XWLC-05,that srparatedly was 100umol/L,50umol/L,10umol/L,1umol/L,0.1umol/L and 0.01umol/L.3,When the concentration of Benzo(a)ypyrene was 100umol/L,there was the inhibitory action to the expression of the Aurora-A protein in the XWLC-05.While when the concentration of Benzo(a)ypyrene was 50umol/L,10umol/L,1umol/L and 0.1umol/L,it might enhance the expression of Aurora-A protein in the XWLC-05.And it played no role in the expression of Aurora-A protein in the XWLC-05 when the concentration of Benzo(a)ypyrene was 0.01umol/L.4,The inhibition role of the Benzo(a)ypyren to the expression of Aurora-A protein in the XWLC-05 was more obviously in the 48th than in the 24th when the concentration was 100umol/L.And the enhancement property of the Benzo(a)ypyren to the expression of Aurora-A protein in the XWLC-05 was more obviously in the 48th than in the 24th when the concentration was 50umol/L,10umol/L,1umol/L and 0.1umol/L.While there was no difference to espression of Aurora-A protein between in the 24th and 48th,when the concentration of Benzo(a)ypyren was 0.01umol/L.Conclusion:1,The level of Aurora-A proteinum expression in the cells of the adenocarcinoma of Xuanwei Lung Cancer-05 was higher.2,There was a "dose-effect" relation between the expression of Aurora-A proteinum and the concentration of Benzo(a)ypyrene in the Xuanwei Lung Cancer-05.That was to say,the expression of Aurora-A protein was stronger when the treatment concentration was lower in some dose range.But the expression of Aurora-A protein become thinner and thinner along with the depressing of the treatment concentration of Benzo(a)ypyrene when the the treatment concentration of Benzo(a)ypyrene cut down to a differential level.And it strengthened along with the time growth.
