| Objective In order to study the mechanism of the Tacrolimus in treating photoallergic dermatitis,we investigate the effects of Tacrolimus on UVA irradiation on HacaT cell,including:1.The proliferation of HacaT cell and the protective effects of Tacrolimus(TM)in vitro.2.The effect of tacrolimus(TM)in the expression of ICAM-1 on the HacaT cell after UVA exposure.3.The effect of tacrolimus(TM)in the expression of VEGF on the HacaT cell after UVA exposure.On the same time,to investigate the efficacy and safety of using 0.1% tarcrolimus ointment in adults with photoallergic dermatitis.Method The proliferation of Hacat cell were observed by MTT method.The expression of ICAM-1 and VEGF on the HacaT cell were measured by ELASI and immunohistochemistry after UVA expressure with different intensity.We enrolled 30 patients,to take oral the tripterygium hypoglaucum hutch tablet,at the same time,one side to topical application of the tacrolimus ointment,the other side,to topical application of the zinc oxide paste.Follow-up visits were scheduled on day 1,2,4 weeks post-treatment.The therapeutic effect and safety were evaluated.Results 1.The proliferation of HacaW cell was increased by UVA irradiation after 48h(P<0.05),TM can inhibited The proliferation of keratinocytes after 48h treatment(P<0.05).2.The group in that the UVA exposure was practiced for 24h had significantly more ICAM-1 expression on HacaT cell that the group without UVA exposure.The use of TM was showed to inhibit significantly this the espression of ICAM-1. 3.The group in that the UVA exposure was practiced for 24h had significantly more VEGF expression on HacaT cell that the group without UVA exposure.The use of TM was showed to inhibit significantly this the espression of VEGF.4.The efficacy rates were 93.3%and 86.6%in patients treated with the 0.1% tarcrolimus ointment and the zinc oxide paste,respectively(P<0.001).Skin physiologic function was evaluated by non-invasive methods(Cutometer).It showed the moisture content and the lipid of cutaneous increase after treatment,but the tacrolimus ointment was better than the zinc oxide paste(P<0.05).the elasticity of skin had not obviously alteration(P>0.05).Skin itching and burning on the application site was a common but temporary side effect.Conclusion(1)TM can inhibit the proliferation of HacaT cell was increased abnormally by UVA irradiation.The use of TM had an inhibitory effect of the expression of ICAM-1 and VEGF on the HacaT cell after the UVA expresure. (2)0.1%tarcrolimus ointment is safe and effective in adults with photoallergic dermatitis. |
