Comparative Study On The Study The Effects Of CTHH And THH On The Mouse's Splenic Lymphocyte Proliferatio,Activation

objective:To study the effects of CTHH and THH on the mouse's splenic lymphocyte proliferatio,activation mark such as CD69 and CD25,cytokine such as IL-4 andγ-IFN;Providing molecular pharmacology evidence for illuminating immunosuppressive action.Methods:Separating the mouse's splenic lymphocyte sterilitily CTHH and THH of different density.The proliferation of the ConA stimulated splenic lymphocyte was measured with MTT;The activetion marks such as CD69 and CD25 are measured with flow cytoeter;cytokines such as IL-4 andγ-IFN are measured with euzyme linked immunosorbent assay.Result:1.The inhibitory action of CTHH and THH on mouse's splenic lymphocyte has evident dependability with density and time,when the cells are cultivated for 72 hours and the CTHH density is over 200ug/ml,when the cells are cultivated for 72 hours and the density is over 120ug/ml,the inhibitory action of CTHH and THH is highly significant.2.CTHH has inhibitory action on the early metaphase activation mark such as CD69,CD25,while THH has inhibitory action on only nonage activation mark such as CD69.3.CTHH and THH have inhibitory action on IFN-γsecreted by Th1 cell and IL-4 secreted by Th2 cell,and the inhibitory action has evident dependability with density.Conclusion:1.CTHH and THH both have inhibitory action on prolixferation splenic lymphocyte,and CTHH is more powerful.2.THH has evident suppression on nonage activation mark such as CD69 while it has no suppression on intermediate stage activation mark such as CD25.CTHH has inhibitory action on both nonage activation mark and intermediate stage activation mark.3.CTHH and THH both have evident suppression on cytokine IL-4 and IFN-γ,and CTHH is more powerful.
