Objective: The purpose of the study is to observe the effect after the application of fluvastatin in early-stage radiation heart injury rat model,to explore effective prevention and treatment on injury induced by radiotherapy.Methods:1. 52 Healthy SD rats(180-220g)were randomly divided into blank(C) group, irradiotherapy (R) group,fluvastatin (F)group.2. The rats of group R,F were injected intraperitoneally by 0.1 ml/kg Chloral Hydrate, after anesthesia they were irradiated with accelerator linear at a dose of 25Gy thoracically. The rate of dose was 2 Gy / min,and the distance was 1 m.Seven days before radiation the rats in F group started to be treated with FLU at a dose of 20mg.kg-1.d-1 by garaging. Then in the second day after radiation FLU was gastrogavaged at a dose of 20mg.kg-1.d-1 and continued to the end of experiment in group F.3. Rats were executed at 5d,15d,30d and 60d after radiation,then a part of heart tissue were observed HE staining,MASSON staining,toluidine blue staining,and transmission electron microscope.4. The contents of ET-1,SOD,MDA and cTnI in blood serum were detected on the day5,15,30,60.Results:1. Group R suffered from myocardial congestion and edema on the day 15,30,60.The area of mesenchyma increased in group R after day 15.Collagenous fiber increased since 30 day in vessel surrounding and under endocardial.It was obvious on the day 60.After 15 d,30d,60d, mast cells increased significantly in R group and F group. The difference was not significant.2. Transmission electron microscope showed that myocardial mitochondrial swell and endothelial cells of capillary were injuried in group R on the day 30,60.Collagenous fiber in myocardial tissues proliferated in group R on the day 60.3. R group cTnI increased more significantly in serum than C group on the day 15d,30d,60d(P<0.05),and F group could decreased it on the day 30(P<0.05).4. R group ET-1 increased more significantly in serum than C group on the day 5d,15d (P<0.05),and F group could decreased it on the day 5d,15d(P<0.05).5. R group MDA increased more significantly in serum than C group on the day 5d (P<0.05),and F group could not decreased it on the day 5d,15d,30d,60d(P>0.05).6. R group SOD increased more significantly in serum than C group on the day 5d,15d (P<0.05),and F group could not decreased it on the day 5d,15d,30d,60d(P>0.05).Conclusions:1. FLU have a protective effect on myocardial cells and vascular endothelial cell injury induced by irradiation.2. FLU could alleviate radiation-induced heart injury by inhibiting the aggregation of inflammatory cell and the formation of collagen fibers.3. ET-1 in serum could be used as a sensitive marker in early radiation-induced heart injury. Serum cTnI could predict the occurrence of radiation myocardial injury.Through the two markers,we find FLU have a treatment on myocardial injury induced by radiotherapy.4. After exposure to radiation,the level of oxidation and antioxidation could enhance,FLU have no effect on it.5. Mast cells increased obviously by the local radiation.The difference was not significant between R group and F group.The role of mast cell in radiation injury and radiation repair is unclear.
