Objective:The important items polycystic ovary syndrome (PCOS) is a common kind of endocrine disorder disease which is the result of internal secretion and sugar metabolism exceptionally among the women with childbearing ages. Excessively are many, the ovulation barrier take the androgen, the insulin resistance, the menstruation detuning merge two-sided bursa ovary increases as the main characteristic, the disease incidency rate is 6%~8%. By the result of PCOS, sterile composition feminine sterility occupied about 10%. The former general scholars studied the occurrence and development of PCOS usually from The Under Cerebral Ganglion - pituitary Gland - germ cells axis's function detuning, obese, aspects and insulin resistance and heredity factor and so on, but the accurate pathogenes is still not clear. In recent years, the domestic and foreign researchers discovered, the disorder of certain ovary independent local control factor was a important link of PCOS's morbidity, the leukemia inhibiting factor (leukemia inhibitory factor, LIF) and the blood vessel bast growth factor (vascular endothelial growth factor, VEGF) are two kinds of important ovary independent local control factors, the expression of the two factors were few reported in the ovary organization. According to related report, the exceptional expression of these two kinds of factors possibility has close correlation to PCOS morbidity.LIF is in IL-6 family, has the different biology activeness in the different organizations and cells. It can adjust the embryo stem cell, the archaeocyte, the liver cell and the endothelial cell and the growth and differentiation of many kinds of cells. In recent years, the massive research indicated that, LIF may affect the reproduction activity, including graafian follicle's growth, embryo growth, differentiation and so on. But the pathogenesis and function of LIF in PCOS is still not extremely clear. VEGF is the most important cell factor which is known as the promotion blood vessel endothelial cell's growth and the strongest blood vessel insightful medicinal preparation, is the most essential regulative factor in the new blood vessel production process. The female reproductive system is all follow the periodic blood vessel production and the degeneration in each reproductive cycle, in recent years, people have been clear about the relations which produce is with blood vessel production special correlation of VEGF and the blood vessel of female reproductive system, but further studies are needed in PCOS pathogenesis.Therefore, this article used the model of the PCOS's rats to observes the expression of LIF and VEGF on ovary organizations by using inununohistochemieal method and discussed the function of LIF and VEGF in pathogenesis of polycystic ovary syndrome.Material and Method:Fouty clearing class SD female rats (24days) with regular estrus cycle were divided into two groups,including experimental group of twenty rats which will be made as animal models and blank control group of twenty rats. All rats are be killd by cutting heads on the 36days. Heart blood was obtained and 2000r/min centrifugal 10min, serum were kept in a freezer at -40℃for subsequent hormone determination including estradiol (E2),luteinizing (LH),follicle stimulating hormone (FSH) and testoterone (T) with radioinununoassay (RIA) to estimate the situation of the endocrine changes; take out the rat's ovaries, after called heavy and records, completely fix by 10% formalin solution and were embedded with paraffin, then makes the paraffin section, detect the expression of the LIF and VEGF of the ovaries by using immuno- histochernieal method.Statistical analysisThe data was calculated using mean±SD. The mean of each group of rats was compared by one-way ANOVA. SPSS 13 was used for studies. P<0.05 was considered significant.Results1.The weight of the ovary of the experimental group is increased obviously. The color of the ovary surface is pale when was seen with own eyes, under the light of the microscope we can see the expanded follicle, the pellet cellular layer changes thin even vanishes, the granular cell arrangement is loose, the membrane and the intersititial cell increase. 2.The LH and T increase obviously in the experimental group but the E2 and FSH decrese(P<0.05).3. The expression intensity of LIF on the granular cell, the follicular membrane cell and the intersititial cell of the experimental group are lower than the control group obviously (P<0.05).4.The expression intensity of VEGF in the granular cell and the sintersititial cell of the experimental group is higher than the control group obviously (P< 0.05), meanwhile on the membrane cell have no significance(P>0.05).Conclusions1. By using progesterone and HCG, the model of PCOS rats can be served as the ideal model of PCOS which was extremely similar to the PCOS changes regardless from the histology change or the hormone level. It has high dependability and is prior to the used PCOS model in our comodels for study of human disease.2. The expression of LIF and VEGF in the ovary close is highly correlated with the originated mechanism of PCOS.
