| Background & Objective:Cell apoptosis plays an important role in the development and progression of malignant tumor.Among apoptosis regulators,Bcl-2 inhibits apoptosis and cause tumors.Bag-1 is also an inhibitor of apoptosis.Encoded by Bag-1 gene which called Bcl-2 associated athanogene-1,Bag-1 protein is a newly discovered Bcl-2 binding protein.But it does not belong to Bcl-2 family.It is a multifunctional protein whose main functions are inhibiting apoptosis with binding to Bcl-2 and restraining cell apoptosis independently.Besides,it adjusts cell's function through many ways to promote the development of tumors.It can also react with cytoskeletal protein,thus promote tumor cell migration.This study was designed to investigate the relationship of Bag-1 and Bcl-2 protein expression in normal gastric mucosa,atypical hyperplasia and carcinoma tissues,and its clinical significance.Methods:The expressions of Bag-1 and Bcl-2 protein in 20 normal gastric mucosa,20 gastric mucosa atypical hyperplasia and 60 gastric carcinoma tissues were examined with immunohistochemical staining.and the analysis of the positive expression was carried out through the imagine analysis system.All of the gastric carcinoma tissues were gastric adenocarcinoma,and were divided into 3 groups according to cell differentiated type.20 normal gastric mucosa was taken trough gastroscope and examined by HE staining.Results:The positive expression rates of Bag-1 protein in normal gastric mucosa,gastric mucosa atypical hyperplasia and gastric carcinoma tissues were 10%,45%and 73.33% respectively.The positive expression rates showed increasing trend from normal gastric mucosa to gastric mucosa atypical hyperplasia to gastric carcinoma tissues,and the difference has statistic meaning(P<0.05).No correlation was found between expression levels of Bag-1 protein and the age and sex of patients,but expression of Bag-1 protein with differentiation degree(P<0.05).The positive rates of Bcl-2 protein in normal gastric mucosa,gastric mucosa atypical hyperplasia and gastric carcinoma tissues were 20%.65%and 68.33%respectively. The positive expression rates of Bcl-2 protein in gastric mucosa atypical hyperplasia and gastric carcinoma tissues were significantly higher than that in normal gastric mucosa tissues(P<0.05). The expressions of Bcl-2 protein were not related to the age,sex of patients and cell differentiation(P>0.05).Bag-1 protein expression was positive correlation with Bcl-2 protein expression in gastric carcinoma(r=0.400,P<0.01).Conclusions:In gastric carcinoma,Bag-1 and Bcl-2 genes are related to cell apoptosis. Compare to Bcl-2,the apoptosis regulator activity of Bag-1 is more dominant.The overexpression of Bag-1 protein plays an important role in the development and progression of gastric carcinoma.The expression levels of Bag-1 protein is related to the cell differentiation of gastric carcinoma and may be used to predict the malignant degree of gastric carcinoma.Bcl-2 protein also plays an important role in the occurrence of colorectal carcinoma.The occurrence and progression of gastric carcinoma are controlled by a variety of genes,and Bcl-2 and Bag-1 protein in the development of colorectal carcinoma enhance each other. |
