Objective: To detect the expression of EPOR in ovarian epithelial tumor. Study the biologic effect of rHuEPO to ovarian epithelial tumor cell HO8910 line and xenografts of ovarian epithelial tumor cell in nude nice.Methods: Ovarian cancer cells HO-8910 were training in the RMPI-1640 medium recovery containing 10% fetal bovine serum at 37℃, 5% CO2 environment . After 2 d stability subculture. Take six bottles of HO-8910, Join rhEPO of 50 U / mL, 100U/mL, 200U/mL to the medium, as the experimental group, 2 bottles of ovarian cancer cells HO-8910 without rhEPO as the control group, after 48 h and 72 h ,RT-PCR analyses were used to detect the expression of EPOR in ovarian epithelial tumor cell,EPOR was detected in ovarian epithelial tumor cell by using reverse transcription polymerase chain reaction ,the relatively uniform levels ofβ-actin transcript expression between the various samples in this study allowed the use ofβ-actin as the standard .After 32cycles PCR products were separated by 1% agarose gels electrophoresis ,and photographed with Imaging system, The relative level of expression of EPORmRNA was therefore computed with respect to the mRNA expression level of the referenceβ-actin transcript . FCM assay was used to delect rHuEPOs effect to ovarian epithelial tumor cell proliferation and inhibition ratio due to Hydroxycanpto- thecine.Results: Compared with the control group, experimental group expressed lower EPOR(P<0.01),(1) The different concentrations of rhEPO in the treated group 48 h and 72h, Compared with the control group, P<0.01; And the different concentrations of the treated group rhEPO EPORmRNA expression of the relative value than the control group decreased. But dealing with different concentrations between groups EPORmRNA expression of no significant difference in the relative value; For the same concentration of EPO-treated group compared at different time points P﹥0.05.(2) The different concentrations of rhEPO in the treated group 48 h and 72h, Compared with the control group, P<0.01; And the different concentrations of EPO treatment group the percentage of cells in S phase compared with the control group decreased。But dealing with different concentrations between groups of cells in S phase percentage of no statistical difference. For the same concentration of EPO-treated group compared at different time points. P﹥0.05. Conclusion: (1) The expression of EPOR was observed in ovarian epithelial tumor cell.(2)Use rHuEPO couldn't promate the cancer,s progression and bring negative effect in clinical practice. S phase cell of HO-8910 have not increased sinnificantlt when incubated for 48h and 72h with different concentrations of rHuEPO . In ovarian epithelial tumor cell have no significant with rHuEPO . therefore R-HuEPO can be used for treatment with the anemia associated with ovarian cancer...
