The PTCH Expression In Intestinal Metaplasia,Atypical Hyperplasia And Carcinoma Of Gastricmucosa

Objective To investigate the relationship of PTCH in Hedgehog signal transduction pathway and the differential stage,invasion,clinical stage,caregogensis of gastric carcinoma..Methods The expression of PTCH proteinum was evaluated by immunohistochemistry in 42 cases of gastric cancer tissues,15 cases of atypical hyperplasia tissues,20 cases of intestinal metaplasia tissues,and 7 cases of normal gastric tissues.To compare the expression of PTCH proteinum in atypical hyperplasia tissues,normal gastric tissues,intestinal metaplasia tissues and gastric cancer tissues.And simultaneously,detecting and semiquantitativing the expression of PTCHmRNA by Rt-PCR in 42 cases of gastric cancer tissues,15 cases of atypical hyperplasia tissues,20 cases of intestinal metaplasia tissues,and 7 cases of normal gastric tissues to investigate the relationship of PTCH and caregogensis of gastric carcinoma.In addition,we compared the expression of PTCH with clinicopathologic characteristics of gastric cancerResult①The expressions of PTCH gene in normal tissues,intestinal metaplasia tissues, atypical hyperplasia tissues and gastric cancer tissues were difference and increased with staging(P＜0.05).PTCH gene was not detected in normal tissues;In intestinal metaplasia tissues,the expression of PTCH proteinum were masccline in 2 pous of intestinal metaplasia tissues(10%),the dyeing intensity were(+),the oppositive expression of PTCHmRNA was 0.07±0.04;In atypical hyperplasia tissues,the expression of PTCH proteinum were masccline in 6 pous(40%),among the total,the dyeing intensity were(+) in 2 pous and( ++) in 4 pous,the oppositive expression of PTCHmRNA was 0.18±0.10;In gastric cancer tissues,the expression of PTCH proteinum were masccline in 33 pous(78.6%) and the dyeing intensity were(+++) and(++++) in 19 pous,the oppositive expression of PTCHmRNA was 0.62±0.11.②The expression of PTCH protein in gastric cancer tissues was correlated closely with differentiated degree,infestation and clinical stage of gastric cancer.Conclusion①PTCH was related with differentiated degree,infestation and clinical stage of gastric cancer.The expression would be of some value to the judgement on prognosis.②PTCH may participate the process of gastric carcinogenesis.
