Standard Trizol protocol was used for RNA isolation from tissues in this study. Labeled cRNA was prepared and hybridized with the Affymetrix human U133 Plus 2.0 oligonucleotide microarray which contains 14,500 genes.We find that there is signifant difference in the gene profile between benign NF and MPNST from the same patient.In MPNST,463 genes were found upregulated in eight-fold or great level while 1215 genes were downregulated.The foemer genes which were upregulated in MPNST included cell cycle,tumor or apoptosis,cell adhesion,motor activity genes,transporter activity,signal transducer activity,and so on.Some of them may be correlated with tumor microinviroment,or acted in immune response.Maybe the uncontrol expression of them help the malignant cells escape from NK cells' immune surveillance in successful oncogenesis.The results are helpful to elucidate the molecular basis and mechanism of the meligancy of the NF1.
