[Objective]To Construct immunocompromised New Zealand rabbit models complicated pneumonomycosis infected with Cryptococcus neoformans,Candida albicans and Aspergillus fumigatus.Provide basement for further research study of clinic,pathology,imageology.[Materials and Methods]We choice 72 healthy male New Zealand rabbits divided into 3 group:group of Cryptococcus neoformans(n=24),group of Candida albicans(n=24) and group of Aspergillus fumigatus(n=24).We divided every group into 2 subset groups experiment animals(n=18)and control animals(n=6).Control animals of Cryptococcus neoformans group don't inject immunosuppressive agent,but inoculate soliquoid of cryptococcus neoformans.Control animals in Candida albicans group and aspergillus fumigatus group are inject immunosuppressive agent,but inoculate tales doses isotonic Na chloride.We use the same method to set up the animal model of immune suppression in animals in Cndida albicans group and experiment animals in cryptococcus neoformans group.In the fist five days,we use cytarabine intravenous injection every day to setup the animal of immue suppression with the dose of 440mg/m2.Then after the 6th day,we use cytarabine intravenous injection every other day to o maintain immue suppression.From 4th day,we use intravenous injection antibiotics to revent the bacterial infection for all rabbits of groups of Candida albicans and cryptococcus neoformans with vancomycin (15mg/kg),ceftazidime(150mg/kg) and Gentamicin(5mg/kg).on 6th day we use the method of per cutem puncturation airtube to intratracheal injection inject bacterium fluid 0.4ml involvement 1×108 cryptococcus neoformans or bacterium fluid 0.2ml involvement 1×108 Candida albicans.We inject Control animals in Candida albicans group with tales doses isotonic Na chloride.In the group of Aspergillus fumigatus,we use cytarabine intravenous injection in th fist five dyas and 8th and 9th days with the dose of 440mg/m2,and radiosone intravenous injection in the fist two days with the dose of 5mg/kg to setup the animal of immue suppression.From 4th day,we use injection antibiotics to revent the bacterial infection for all rabbits of the group with vancomycin (15mg/kg),ceftazidime(150mg/kg) and Gentamicin(5mg/kg).On second day we use the method of per cutem puncturation airtube to intratracheal injection bacterium fluid 0.4ml involvement 1×108 Aspergillus fumigatus.We inject Control animals of group of Candida albicans tales doses isotonic Na chloride.We draw venous blood from the fringe of ear for blood routine examination to get the message of granulocyte on the fist day of experiment and the day of inoculation.Thoracic CT scan was performed before inoculation and at 2,4,6,8,10,12,14d after inoculation respectively.We execute 3~4 rabbit that have abnormal manifestation in Thoracic CT imagine every 2~3 days,the quantity will decrease if there are some rabbit died,then abstract the animals lung,and get specimens for the examinations of mycetology and pathology to comprehend the growth of mycetes and pathogenetic condition.[Results]1.The amount of granulocyte obviously decreased after intravenous injection of immunodepressant.The discrepancy in two group of numerus has statistical significance.2.Achievement ratio of innoculation in Cryptococcus neoformans experiment group is 100%(18/18),Achievement ratio of modeling is 83.33%(15/18).Achievement ratio of modeling in control group is 33.33%(2/6),there is obvious statistical discrepancy in two group.3.Achievement ratio of innoculation in Candida albicans experiment group is 100% (18/18),Achievement ratio of modeling is 88.89%(16/18).Achievement ratio of modeling in control group is 16.67%(1/6),there is obvious statistical discrepancy in two group.4.Achievement ratio of innoculation m Aspergillus fumigatus experiment group is 100%(18/18),Achievement ratio of modeling is 88.89%(16/18).Achievement ratio of modeling and innoculation in control group is 0%(0/6)。 5.Abnormal manifestation in Thoracic CT imagine include GGO,air-space consolidation,and usually associated with other findings including bronchovasular bundle thickening,nodules and reticular or linear opacity.[conclusion]Useing the method of per cutem puncturation airtube to intratracheal injection and regular thoracic CT scan,we can Construct the models of pneumonomycosis fitting study of imageology by injecting soliquoid of Cryptococcus neoformans,Candida albicans and Aspergillus fumigatus into lungs of immunocompromised New Zealand rabbit. achievement ratio of the method is high.The operation of the method is convenient.The reproducibility is better.In the experiments,the model of rabbit pulmonay Cryptococcus have had no report.The models of pneumonomycosis in immunocompromised New Zealand rabbit respectively are construted successfully which provid valuable tool for control study of radiology and pathology.
