Objective To prepare the mouse forestomach carcinoma vaccines based on dendritc cells(DCs) from mouse bone marrow monocytes induced by OK-432 combined with freeze-thaw tumor cells lysates and to observe its anti-tumor effect in vivo so as to evaluate the applied value in gastric cancer immunotherapy for the clinical treatment later.Methods Mouse bone marrow monocytes(BMMCs) were isolated by adherence method and cultured in RPMI1640 containing 10% FCS, rmGM-CSF and rmIL-4 and then cells were loaded with freeze-thaw cell lysates of mouse forestomach carcinoma and stimulated by OK-432 so as to control the maturation of DCs in vivo for the preparation of gastric cancer vaccines. Cells were observed by the optical microscope and the electron microscope. Method of flow cytometry were used to determined cells surface antigens. DCs vaccines were injected into the forestomach carcinoma bearing mice through the caudal vein in order to observe the immunotherapy capacity.Results Cells appeared typical morphology of DCs and express more antigen CD83 and CD86 on the surface induced by OK-432 combined with freeze-thaw tumor cell lysates and showed more significant tumor-inhibitory effect in vivo than that in other groups.Conclusion Mouse bone marrow monocytes(BMMCs) were isolated by adherence method and induced by GM-CSF combined with IL-4 and then were impulsed with OK-432 combined with freeze-thaw tumor cell lysates, which can abundantly amplify maturation DCs and effectively inhibit the growth of tumor in bearing cancer mice. So it would be a kind of fit for clinical gastric cancer immunotherapy with further reaserches and lay the foundation of preparation of dendritic-cells-based vaccines for the other cancers.
