Expression Of HIF-1α,VEGF In Villus Of Unexplained Recurrent Spontaneous Abortion And Its Relation With MVD

Objecive:To examine the expression of HIF-1αand VEGF in villus of patients with uncxplained recurrent spontaneous abortion(URSA),and to study its relation with MVD.Then to detect the effect of hypoxia and HIF-1αon the expression of VEGF in the conditional cultured cytotrophoblasts would provide guidance for the study of the pathogenesis of URSA.Method:Immunohistochemical SP method was used to examine the expression of HIF-1α,VEGF and MVD in villus of 28 URSA patients and 20 healthy pregnant women.The result was analyzed by SPSS11.5.Result:1)Villi edema,cytotrophoblast cell hyperplasia and syncytiotrophoblast cell decrease were observed in the case group.Cytotrophoblast edge sharpness,cellular nucles vascuolar,syncytiotrophoblast comparatively large to confluence one another were observed in the control group.2)HIF-1αand VEGF were observed chiefly in plasma and membrance of cytotrophoblasts cell and vascular endothelial cell both in the two groups.And the mesenchymal has little stain.However the expression degree of HIF-1αand VEGF in the villi of the case group were decreased significantly than that of the control group.3)Angiodysplasia,lumina narrow or no apparente lumina,little blood cell in the lumina of the case group,the verage of MVD was significant decreased in the case group than that of the control group.4)Positive correlation was found between HIF-1αand VEGF.Positive correlation was found between HIF-1α,VEGF and MVD.Conclusion:1)Compared to the villi of normal pregnant,there were recessive changs in the villi of the URSA.2)HIF-1αand VEGF expression of the villus may play an important role in the normal pregnant,possibly through affecting the cytotrophoblastic invasion and placental vascular reconstruction via the modulation of VEGF gene transcription.3)The expression levels of HIF-1αand VEGF in villus were lower in URSA group than those in control,and the villus were angiodysplasia.It was indicate that HIF-1αcan regulate VEGF by resuming the oxygen pressure.These would result the URSA.4)The resesrch of the expression of HIF-1α,VEGF and MVD in villi and the correlation between them might offer theory proof for prevention and treatment of URSA.
