| Objective:To investigate the presence of drug resistance mutation on protease and reverse transcriptase among drug naive human immunodeficiency virus type 1 infected adults and children patients in Hunan province of China.Methods:Viral RNA samples were collected from the sera of Human Immunodeficiency Virus type 1(HIV-1)infected patients without receiving highly active anti-retroviral therapy(HAART).and 184 pol gene of HIV-1 were amplified by reverse transcriptase polymerase chain reaction(RT-PCR),then the amplified fragments were sequenced and analyzed.Result:Analyze the 120 purpose gene fragment,the PI minor mutations were generally existed,including major types of M36I,H69K, I13V and I93L.The accumulation can also cause resistance on protease inhibitor.The resistance mutations on reverse transcriptase were rare. There are two samples that had many mutations on both of nucleoside reverse transcriptase inhibitor(NRTI)and non-nucleoside reverse transcriptase inhibitor(NNRTI)simultaneously.Conclusion:There were primarily drug resistance in naive HIV-1 infected adults and children patients in Hunan province,but the prevalence was low,termly and systematic detections for gene resistance will be necessary before and during HAART treatment to guide anti-HIV therapy.This study initial discovered that the regularity of drug resistance mutations on children HIV-1 infected patients is similar as that of adults'. |
