| [Objective]:To investigate the relationship between the level of Foxp3 mRNA in the peripheral blood monocytes of kidney transplantation patients receiving different immunosuppression programs,and provid references for the selections of clinical drugs after kidney transplantation.[Methods]:30 cases kidney transplantation patients were selected and were divided into two groups:CNIS+MMF+Pred group(A group, 19 cases)and RAPA+MMF+Pred group(B group,11 cases)according to different immunodepressive programs.Among the total,A group was divided into two subsets:CSA+MMF+Pred group(A1 group,10 cases) and Tac+MMF+Pred group(A2 group,9 cases);B group was also divided into two subsets:RAPA+MMF+Pred group after received CSA+MMF+Pred group(B1 group,6 cases)and RAPA+MMF+Pred group in full range(B2 group,5 cases).Simultaneously 8 cases urinaemia patients(end-stage renal disease group,C group)and 5 cases healthy men(normal control group,D group)were selected to be the controls.Real time quantitative polymerase chain reaction(real time PCR) was used to examine Foxp3 mRNA expression in the PBMCs from the cases of four groups and Simultaneously their creatinines were also detected,then we compared the differences of four groups expressions and relations to immunodepressive programs.[RESULTS]:1.Foxp3 mRNA expression was significantly lower in patients receiving CNIS+MMF+Pred group(0.1089±0.09672)than the normal control group(0.9380±0.36141),the end-stage renal disease group(1.1200±0.57029)and RAPA+MMF+Pred group(1.1264± 0.65645).(P<0.01)2 The level of Foxp3 mRNA in the PBMCs among RAPA+ MMF+Pred group(1.1264±0.65645),the normal control group(0.9380±0.36141)and end-stage renal disease group(1.1200±0.57029)were not significantly different.(P>0.05)3 The level of Foxp3 mRNA in the PBMCs between CSA+MMF+Pred group(0.1110±0.11010)and Tac+MMF+Pred group (0.1067±0.08602)was also not significantly different.(P>0.05)4 The level of Foxp3 mRNA in the PBMCs between RAPA+ MMF+Pred after received CSA+MMF+Pred group(1.3500±0.82835) and RAPA+ MMF+Pred group in full range(0.8580±0.23360)was not significantly different.(P>0.05)5 The level of creatinine in the PBMCs between RAPA+MMF+Pred group and CNIS+MMF+Pred group was not significantly different.(P>0.05)[CONCLUSION]:1 Foxp3 mRNA expression was significantly higher in patients receiving RAPA+ MMF+Pred than the patients receiving CNIS+MMF+Pred.2 RAPA maybe contributed to the up-regulation of the level of FOXP3 mRNA expression in peripheral blood monocytes, which suggested that RAPA maybe play a more important role than CNIS. |
