Objective:To study the effect of adolescent alcohol stimulation on the spatial learning and memory function and the expression of synaptophysin in the hippocampal formation neurons of adult rats.Method:Adolescent SD rats were divided into two groups:saline or alcohol(2g/kg),20 rats in each group.Rats of alcohol group were administered ethanol by intraperitional injection,and the process was repeated daily for 5 continuous days,twice everyday,while rats of control group were received isovolumetric saline injection on the same time,then all of rats were raised normally.When they maturated,rats were tested in Morris water maze for 3 days,and recorded a video of every animal behavior,then the result of Morris water maze was analyzed by using software SPSS 11.5.The hippocampal formation of every rat was tested immunohistochemistry of synaptophysin.Result:With the times of training increased,the escape latency of rats of each group were shortened.Significanted differences of escape latency were found between control and alcohol group(P<0.05),the escape latency of alcohol group is longer than that of control group.The expression of the immunohistochemistry of synaptophysin:The positive immunoproduct of synaptophysin mainly express in the CA3 and gyrus dentatus of the hippocampal formation.They were brown drops and arranged a laminar shape.There is a significant difference between control and alcohol group of synaptophysin content in the hippocampal formation(P<0.05).Conclusion:The expression of synaptophysin was decreased in CA3 and gyrus dentatus of the hippocampal formation of adult rats which had ever received alcohol stimulation in adolescent,the escape latency in Morris Water maze was prolonged,and the capability of the spacial learning and memory was decreased.
