We detected the expression of Cdt1 and Geminin mRNA by RT-polymerase chain reaction(RT-PCR). The outputs are semiquantitative analysised. The Cdt1 and Geminin proteins are detect by flow cytometry . The expression of proteins are quantitative analysised by mean fluorescence intensity.Resultsâ‘ Cdt1 mRNA and Geminin mRNA are all highlier expressed in research group than control group (P<0.05) .â‘¡Cdt1 proteins and Geminin proteins are all highlier expressed in research group than control group (P< 0.05)Conclusionsâ‘ The over-expression of Cdt1 in in Jurkat cells(human T-lymphocytic leukemic cell lines) may be releated to the maligmant proliferation of leukemic cells.â‘¡The over-expression of Geminin in Jurkat cells(human T-lymphocytic leukemic cell lines) may be involved in the maligmant proliferation of leukemic cells.Section II The expression of Cdt1 and Geminin in every phaseof cell cycle in K562 cellObjective To explore the expression of DNA replication licensing factor Cdt1 and the DNA replication inhibitor Geminin in every phase of cell cycle of leukemic cell.Methods Cultivate leukemic cell K562 in vitro. K562 cell is synchronized by thymidine.The efficiency of synchronization is detected by flow-cytonetry(dying by PI).The Cdt1 and Geminin proteins are detected by flow cytometry .
