Objective:The aim of this study is to examine the expression of TLR2 and TLR4 in the peripheral blood mo nouclear cells of SLE and infection diseases, and explore their association with disease stage and clinical manifestations. In order to explore their association with such diseases.Methods:we chose 36 SLE patients (19first treatment patients) and 17 bactelial infection patients and 15 healthy controls. We compared the expression of TLR2 and TLR4 in peripheral blood monouclear cells among SLE of first treatment group,infection group and hleathy control group(12 individuals) by flow cytometry. Then according to SLEDAI score, we divided 36 SLE patients into 2 groups: active group (19 individuals), non-active group(17 individuals), and compared the expression of TLR2 and TLR4 between two groups. According to infection, we divided SLE active group into SLE active with infection group and SLE active without infection group, and SLE non-active group into SLE non-active with infection group and SLE non-active without infection group. We analyzed the correlation between TLR2/TLR4 and ESR, CRP, C3, C4, the amount of anti-double strands DNA, SLEDAI score and urine protein excretion.Results:1. The expression of TLR2/TLR4 andTLR2/TLR4 MIF in bacterial infection group was significantly higher than the normal group(P<0.005).2. The expression of TLR2/TLR4 andTLR2/TLR4 MIF in SLE first-treatment group was significantly higher than the normal group(P<0.005).3. The expression of TLR4 and TLR4 MIF in SLE first-treatment group was significantly higher than bacterial infection patients(P<0.005).4. The expression of TLR2 and TLR2 MIF between SLE active group and SLE non-active group were no difference among such disease groups(P>0.05). The expression of TLR2/TLR4 andTLR2/TLR4 MIF had on difference between SLE active with infection group and SLE active without infection group (P>0.05); The expression of TLR2/TLR4 andTLR2/TLR4 MIF had on difference between SLE non-active with infection group and SLE non-active without infection group (P>0.05) .5. The expression of TLR2 had correlation with CRP and C4,and TLR2MIF had correlation with C3.While TLR4 and TLR4 MIF had no correlation with ESR, CRP, C3, C4, the amount of anti-double strands DNA and SLEDAI score.Conclusion:1. The expression of TLR2 and TLR4 were higher between SLE first-treatment group and bacterial infection group than in normal group. Which indicated that TLR2 and TLR4 have correlation with SLE and infection diseases.2. The expression of TLR4 and TLR4 MIF in SLE first-treatment group was significantly higher than the bacterial infectionl group.Which indicated that TLR4 had further correlation with SLE pathogenesis.3. The expression of TLR4 between SLE active and non-active group had no statistical difference,and TLR4 had correlation with CRP and SLEDAI and so on,which indicated that TLR4 had no correlation with SLE SLEDAI.4. The expression of TLR2 and TLR4 between SLE active with infection and non-infection group had no statistical difference. Which indicated that infection the expression of TLR2 and TLR4 in SLE active group,and further indicated the expression of TLR2 and TLR4 had correlation with SLE itself.
