Object: To detect the methylation status of MGMT gene's promoter in Uigur cervical squamous cell carcinomas tissues and normal Uigur cervical tissues, to explore the correlation between the methylation status of MGMT gene's promoter and Uigur cervical squamous cell carcinomas as well as its histopathological grading,clinical stage and lymphatic metastasis. It also will helpful to cervical cancer's individuality therapy. To detect the methylation status of MGMT gene's promoter in plasma of Uigur cervical cancle and normal Uigur cervical,finding a simple and harmless diagnosis method in cervical cancer.Methods: Collect Uigur cervical squamous cell carcinomas tissues speciments (frozen tissue 61 cases, paraffin-embedded tissues 25 cases) and normal Uigur cancer tissues speciments(38 cases)from 2005.1 to 2007.6. Collect Uigur cervical cancer plasma speciments (41 cases)and normal Uigur cervica plasma speciments (40 cases) from 2007.1 to 2007.9.Detect the aberrant methylation of MGMT gene by methylation-specific polymerase chain reaction(MSP) .Detect transcription level of MGMTgene in cervical squamous cell carcinomas tissues and normal Uigur cancer tissues by semi-quantitative RT-PCR.Results:(1)Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas tissues was 31%(27/86), Methylation rate of MGMT gene in normal Uigur cervical was 0%(0/38), There was significant difference(P<0.005). Methylation rate of MGMT gene in Uigur Cervical cancer plasma was 26%(11/43), Methylation rate of MGMT gene in nomal Uigur plasma was 0%(0/40).There was significant difference(P<0.005).(2)Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas frozen tissue was 31%(19/61), Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas paraffin-embedded tissues was 32% (8/25). Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas plasma was 26%(11/43), There was not significant difference(P>0.05).(3)Methylation rate of MGMT gene in Uigur cervical well differentiated squamous cell carcinoma is 20%(1/5), Methylation rate of MGMT gene in Uigur cervical moderately differentiated squamous cell carcinoma is 38%(6/16), Methylation rate of MGMT gene in Uigur cervical poor differentiated squamous cell carcinoma is 33%(4/12), There was not significant difference(P>0.05).(4)Methylation rate of MGMT gene in the low level in clinical stage of Uigur cervical squamous cell carcinomas tissues (Carcinoma in situ and I) was (15%,2/13), it was lower than Methylation rate (36%,17/47) of MGMT gene in the high level in clinical stage of Uigur cervical squamous cell carcinomas tissues (II andâ…¢), But there was not significant difference(P>0.05).(5)Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas tissues with lymphatic metastasis is 43%(3/7), it was higher than Methylation rate (36%,17/47) of MGMT gene in Uigur cervical squamous cell carcinomas tissues without lymphatic metastasis. But there was not significant difference(P>0.05).(6)The methylation status of MGMT gene's promoter in Uigur cervical squamous cell carcinomas tissues can inhibit gene's transcription.Conclusion: (1)The methylation status of MGMT gene's promoter partly involves in tumorigenesis of Uigur cervical squamous cell carcinomas.but it hasn't relativity with tumor's histopathological grading,clinical stage and lymphatic metastasis.(2)Methylation rate of MGMT gene in Uigur cervical squamous cell carcinomas plasma is similar to cervical squamous cell carcinomas tissue,it suggest detect promoter methlytion of MGMT gene in plasma can be a simple and harmless diagnosis method in cervical cancer.
